Abstract: The work described in this dissertation is a contribution to a financed research project between the University of Coimbra and a pharmaceutical company, which aims to design an injectable in situ forming drug delivery system for controlled and prolonged drug release. The primary goal of this work is the development of a hydrogel, part of that same system, from polysaccharides crosslinked by Michael addition. This objective can be further divided into specific steps to reach this goal, namely: (i) functionalization of polysaccharides with double bonds and thiol groups, which are necessary for the Michael addition reaction, and characterization of the products; (ii) optimization of the modification reactions to achieve suitable degrees of modification, and (iii) study of the gelation reactions to have hydrogels in the shortest possible time.Hyaluronic acid and carboxymethylcellulose were successfully modified, however, the modification of dextran was also attempted but no positive results were obtained.For the unsaturated carbon-carbon bond introduction, the maximum degrees of substitution were of 19.3 % acrylation of hyaluronic acid with acrylic acid, 45.7 % vinyl sulfonation of hyaluronic acid with divinyl sulfone and 6 % methacrylation of hyaluronic acid with methacrylic anhydride. As for carboxymethylcellulose, a level of 23.5 % vinyl sulfonation with divinyl sulfone was reached.The thiolation of hyaluronic acid achieved at most 8.3 % modification with cysteamine and 11.0 % with cystamine. Carboxymethylcellulose was also reacted with cystamine, resulting in a 28.2 % degree of substitution.Several hydrogels were produced, based in the above-mentioned modified polysaccharides. The thiolation products were not very successful in producing hydrogels. In fact, only one hydrogel successfully transitioned from solution to gel with a thiolated derivative, acrylated hyaluronic acid and dithiothreitol.
O trabalho descrito nesta dissertação é uma contribuição para um projeto de investigação financiado entre a Universidade de Coimbra e uma empresa farmacêutica, cujo objetivo é conceber um sistema de libertação injetável com formação in situ para a libertação controlada e prolongada de fármacos. A principal finalidade do trabalho é o desenvolvimento de um hidrogel, parte desse mesmo sistema, à base de polissacarídeos reticulados por adição de Michael. Este objetivo pode ser subdividido em passos específicos para o alcançar, nomeadamente: (i) funcionalização de polissacarídeos com ligações duplas e grupos tióis, que são necessários para a reação de adição de Michael, e a caracterização dos produtos; (ii) otimização das reações de modificação para atingir graus de modificação apropriados, e (iii) estudo das reações de gelificação para obter hidrogéis num espaço de tempo tão curto quanto possível.O ácido hialurónico e a carboximetilcelulose foram modificados com sucesso, no entanto, também se tentou substituir o dextrano, mas sem sucesso.Para a introdução de ligações carbono-carbono insaturadas, os graus de substituição máximos foram de 19.3 % de acrilação do ácido hialurónico com ácido acrílico, 45.7 % de vinil sulfonação do ácido hialurónico com divinil sulfona e 6 % de metacrilação do ácido hialurónico com anidrido metacrílico. Para a carboximetilcelulose, atingiu-se 23.5 % de substituição com divinil sulfona.A tiolação do ácido hialurónico conseguida foi de 8.3 % com cisteamina e 11.0 % com cistamina. A carboximetilcelulose também reagiu com a cisteamina, resultando em 28.2 % de substituição.Vários hidrogéis foram produzidos com base nos polissacarídeos modificados suprarreferidos. A preparação de hidrogéis a partir de polímeros tiolados não foi bem-sucedida. De facto, apenas houve transição sol-gel de uma mistura de um derivado tiolado, ácido hialurónico acrilado e ditiotreitol.
