Abstract: A tau é uma proteína associada aos microtúbulos e a agregação da tau cuja conformação se encontra alterada origina tranças neurofibrilares (TNFs), sendo estas características da Doença de Alzheimer (DA). A tau é reconhecida por estar num estado hiperfosforilado quando agregada em filamentos helicoidais emparelhados (FHEs), que compõem os TNFs. O papel da proteína tau na DA tem recebido mais atenção ao longo dos anos, devido à crescente necessidade de compreender as implicações do comprometimento dessa proteína no cérebro. Vários esforços foram feitos na tentativa de desenvolver modelos que pudessem providenciar informação acerca dos mecanismos subjacentes à agregação da tau. Além disso, a proteína tau tem sido, mais recentemente, alvo de investigação direccionada ao desenvolvimento de terapias que visam impedir a progressão da doença ou até a formação da patologia da DA. Estas terapias têm como objectivo reduzir os níveis de tau, por exemplo, pela remoção da tau ou diminuindo a agregação da tau. Uma terapia emergente é o uso de uma quimera direccionada à proteólise (PROTAC), que consiste num peptídeo com dois domínios. Um deles liga-se à proteína de interesse e o outro liga-se a uma E3 ligase específica, levando a proteína de interesse à degradação pelo proteassoma.O objectivo deste projeto foi amplificar AD-tau seeds capazes de induzir agregação de endógena wild-type (WT) tau, com o objetivo de desenvolver um modelo de agregação para DA esporádica. Este modelo forneceria ferramentas para estudar o mecanismo de agregação de endógena WT tau em neurónios. Anteriormente, o nosso laboratório desenvolveu um modelo de agregação de tau que expressa full-length tau com a mutação P301L. Esta linha celular foi seeded com fibrilas de tau que tem como fonte de origem pacientes com DA ou com fibrilas de tau recombinante (K18-P301L). Desta forma, duas novas linhas celulares foram desenvolvidas, ADT-1 e Cl.44, respectivamente. Ambas demonstram a capacidade de ter tau agregada e transmiti-la às gerações seguintes à medida que as células se dividem. Purificamos, caracterizamos e avaliamos a potência das seeds extraídas das linhas celulares ADT-1 e Cl.44 em diferentes modelos. Apenas as seeds do Cl.44 demonstram capacidade de induzir agregação da tau, especificamente em células cuja tau tem a mutação P301L. ADT-1 seeds não demonstraram capacidade para induzir agregação em diferentes modelos, permitindo concluir que a amplificação das seeds não teve sucesso.Além disso, testamos o efeito de um PROTAC (TH006) na sua capacidade de diminuir os níveis de tau em diferentes modelos, como uma futura terapia para tratar a patologia da tau na DA. Embora a sua eficácia tenha sido demonstrada num artigo, não conseguimos reproduzir os resultados obtidos, demonstrando que nos nossos modelos o PROTAC é incapaz de levar tau à degradação pelo proteossoma.
Tau is a microtubule-associated protein and the aggregation of its misfolded form into neurofibrillary tangles (NFTs) is one of the hallmarks of Alzheimer’s Disease (AD). Tau is known to be in an hyperphosphorylated state when aggregated in paired helical filaments (PHFs), which compose NFTs. The role of tau protein in AD has been receiving increased attention throughout the years, as there is a growing need to better understand the implications of the impairment of this protein in the diseased brain. A lot of effort has been put in trying to develop models that could give a better understanding on the mechanisms underlying tau aggregation. Moreover, tau protein has also been more recently the target of many therapies research that aim at stopping the progression of the disease or even the development of the pathology in AD. These therapies goal is to reduce tau levels for example by promoting its clearance or by decreasing its aggregation. One emerging proposed therapy is the use of a proteolysis targeting chimera (PROTAC), which consists of a peptide with two domains. One of them binds to the protein of interest and the other binds to a specific E3 ligase, leading the protein of interest to degradation by the proteasome system.The goal of this project was to amplify AD-tau seeds capable of inducing endogenous wildtype (WT) tau aggregation in order to develop a reproducible tau aggregation model as seen in sporadic AD. This model would also provide tools for studying the mechanism of aggregation of endogenous WT tau in neurons. Our lab has previously developed a tau aggregation model that expresses full-length tau bearing P301L mutation. This cell line was seeded with human-derived tau AD fibrils or with recombinant K18-P301L fibrils. Two new cell lines were originated, ADT-1 and Cl.44, respectively. Both demonstrate the ability to aggregate tau and pass it on to the next generations as cells divide. We purified, characterised and evaluated the seeding potency of seeds extracted from both ADT-1 and Cl.44 in different models. Only seeds from Cl.44 appear to be able to induce tau aggregation, specifically cells in which tau bears the P301L mutation. ADT-1 seeds do not aggregate tau in different models, thus proving that the amplification of AD-tau was not successful.Furthermore, we tested the effect of a PROTAC (TH006) in its ability to decrease tau levels in different models, as a future therapy for tau pathology in AD. Although its efficacy was shown in a paper we were not able to reproduce the results obtained, thus in our models the PROTAC is unable to lead tau to degradation by the proteasome.
