Abstract: The influenza viruses, belonging to the Orthomyxoviridae family, are classified into four genus, A, B, C and D, but only type A and B are associated with severe human pathologies. In the human population circulate currently the H1N1 and H3N2 subtypes of the influenza A virus and two lineages antigenically distinct of the influenza B, Yagamata and Victoria. Influenza viruses are responsible for seasonal epidemics and sporadic pandemics of acute respiratory disease, with significant impact on public health (such as high mortality and morbidity, mainly associated with elderly population) and on the economys of societies. Vaccination is currently the most effective way of prevent this infection, but it has some limitations, such as the induction of a short-term and basically humoral immunological response. It also decreases or even nullifies its efficiency caused by a phenomenon known as antigenic variability (antigenic drift and antigenic shift, respectively). Finally, the long period of time for the vaccine manufacturing process compromises its practical usefulness. Faced these handicaps, the scientific community has searched to develop a universal vaccine, through different approaches, that induce a total and long lasting immune protection, available to the population, according specific needs. The approaches are focussed on viral antigens with conserved epitopes, like the domain of the stalk hemagglutinin, the ectodomain of the matrix protein 2 and internal proteins (nucleoprotein and matrix protein 1).
Pertencentes à família Orthomyxoviridae, os vírus influenza classificam-se em quatro géneros, A, B, C e D, sendo que apenas os tipos A e B estão associados a patologia humana severa. Na população humana atualmente circulam os subtipos H1N1 e H3N2 do vírus influenza A e duas linhagens antigenicamente distintas do vírus influenza B, Yagamata e Victoria. Os vírus influenza são responsáveis por epidemias sazonais e por pandemias esporádicas de doença respiratória aguda, com impacto significativo na saúde pública (mortalidade e morbilidade elevadas, maioritariamente associadas à população idosa) e na economia das sociedades. Atualmente, a vacinação representa o meio mais efetivo para prevenir esta infeção, porém apresenta algumas limitações, tais como a indução de uma resposta imunológica de curta duração e fundamentalmente de natureza humoral, a diminuição ou até abolição da sua eficácia pela ocorrência de um fenómeno designado de variabilidade antigénica (antigenic drift e antigenic shift, respetivamente) e por fim o extenso período de tempo inerente ao processo de fabrico da vacina que põe em causa a disponibilização da mesma em tempo útil. Face a estes handicaps, a comunidade científica tem procurado desenvolver através de diferentes abordagens, uma vacina universal que induza uma total e duradoura proteção imunitária, que possa ser disponibilizada de acordo com as necessidades da população. Estas abordagens centram-se em antigénios virais dotados de epítopos conservados, nomeadamente o domínio da haste da hemaglutinina, o ectodomínio da proteína da matriz 2 e as proteínas internas (nucleoproteína e proteína da matriz 1).
