A comunidade científica tem-se preocupado em desenvolver novas estratégias de combate a
esta doença, focando-se na pesquisa de novos alvos terapêuticos e na descoberta de
biomarcadores ideais que permitam uma deteção precoce da doença. Os microRNAs
(miRNAs), moléculas endógenas, com capacidade de regular a expressão genética, têm sido
alvo de inúmeros estudos, desde a sua descoberta em 1993, e têm demonstrado ser
potenciais candidatos a satisfazer estes requisitos. Para tirar partido da totalidade do
potencial destas moléculas é essencial compreender o seu papel nos processos fisiológicos e
as implicações associadas à sua disfunção e desregulação. De facto, a identificação de perfis
de expressão que permitem diferenciar as neoplasias malignas, tem merecido destaque pelo
seu papel promissor no diagnóstico e prognóstico destas patologias. Para além disso,
começam a ser concluídos estudos sobre a segurança e eficácia do seu uso na terapêutica
oncológica. Assim, neste trabalho, discutem-se os principais avanços já alcançados nesta área,
bem como os maiores desafios que ainda têm de ser ultrapassados. Throughout the world, the number of cancer cases has been rising considerably. The scientific community has been concerned in developing new strategies to fight this disease by focusing on the search of new therapeutic targets and on the discovery of ideal biomarkers that allow an early detection of the disease. The miRNA, endogenous molecules that can regulate the genetic expression, have been the subject of innumerous studies since its discovery in 1993 and have been shown as potential candidates to meet these requirements. To take advantage of the full potential of these molecules is essential to understand their role in physiological processes and the implications associated with its dysfunction and deregulation. In fact, the identification of expression profiles to differentiate between malignant neoplasms has been highlighted by its promising role in the diagnosis and prognosis of this pathologies. In addition, studies on the safety and efficacy of their use in cancer therapy are being completed. Thus, this paper discusses the major advances already achieved in this area, as well as the major challenges that still are to be overcome. |
