Abstract: O envelhecimento demográfico resulta da diminuição da taxa de natalidade a par do aumento da esperança média de vida. Evidentemente, a situação sem precedentes que estamos a viver presentemente representa um desafio no sentido de atenuar a morbilidade nos idosos, retardando, tratando ou curando as doenças sempre que possível. Uma das respostas é a terapia celular, particularmente a baseada em células estaminais mesenquimatosas (MSCs) – dada a sua acessibilidade, facilidade de cultura e versatilidade. Este trabalho pretendeu localizar-nos no estado da arte dos estudos com MSCs no âmbito de algumas doenças do envelhecimento, salientar as suas problemáticas mais importantes e eventuais limitações. Foram introduzidas as potencialidades das tecnologias emergentes, como os nanotubos, o pré-condicionamento e a terapia génica. Para a pesquisa bibliográfica utilizaram-se os termos “mesenchymal+stem+cells”, “bone+marrow+stromal+cells”, “multipotent+stem+cells” e “multipotent+stromal+cells”, na base de dados PubMed. Embora numa etapa preliminar de investigação, o uso das MSCs nas doenças do envelhecimento apresenta-se promissor, principalmente com base na aplicação das suas propriedades de tropismo para locais de lesão, imunomodulação e libertação de factores bioactivos. Hoje já existem alguns trabalhos impressionantes, ao nível da clínica, protagonizados pelas MSCs aplicadas à doença crónica. Gerou-se, portanto, uma grande expectativa no sentido de melhorar, ou quem sabe, reescrever a história natural das doenças do envelhecimento.
The current demographic ageing results from the decreasing birth rate in addition to the increasing of average life expectancy. Evidently, this unprecedented situation represents a challenge in lessening the morbility in old age, postponing, treating or curing illnesses whenever possible. One such path is Cellular Therapy, particularly the one based on mesenchymal stem cells (MSCs) given its accessibility, easiness of cell culture and versatility. The purpose of this essay was to point out where we are in regard to the state of the art on the studies with MSCs concerning ageing diseases, underlining its most important problematics and eventual limitations. Focus was given to the potential of emerging technologies such as nanotubes, preconditioning and gene therapy. For the bibliographic literature, the searched keywords were “mesenchymal+stem+cells”, “bone+marrow+stromal+cells”,“multipotent+stem+cells”and“multipotent+stromal+cells” within the PubMed database. Even though they are at a preliminary investigation stage, the use of MSCs on ageing diseases has shown promise, especially when it comes to its tropism properties, immunomodulation and the releasing of bioactive factors. There are already some impressive reports concerning clinical results of the applications of MSCs on chronic diseases. A great expectancy has arisen on the pursuit to improve or, perhaps, rewrite the natural progression of ageing diseases.
