Abstract: A maior parte das leveduras e dos fungos filamentosos, quando em situações de agressão osmótica, acumulam, como substâncias osmorregu-ladoras, polióis. A acumulação intracelular destes alcoóis está relacionada com a fase de crescimento e é proporcional à concentração externa de solutos osmoticamente activos, iónicos ou não iónicos. Nas leveduras Debaryomyces hansenii, Candida famata e Pichia farinosa, o glicerol é a substância osmorreguladora (soluto compatível) principal. Em meios em que é usada glicose, como fonte de carbono, as concentrações mais elevadas deste poliol ocorrem durante a fase exponencial de crescimento. O arabitol é acumulado em concentrações significativas, apenas durante a fase estacionária. Nestas leveduras a substituição da glicose por eritritol nos meios de cultura com NaCl 1 M conduz à acumulação deste poliol, durante a fase exponencial, em níveis comparáveis aos do glicerol em meio com glicose. Este facto demonstra que, nestas leveduras, o glicerol pode ser substituido nas suas funções de soluto compatível, por eritritol, quando este poliol é fornecido exogenamente. No fungo leveduriforme Geotrichum candidum, é o arabitol que desempenha o papel de soluto compatível principal, sendo acumulado durante a fase exponencial de crescimento em meios com glicose, em resposta à diminuição da actividade da água no meio. Durante a fase estacionária, observa-se a acumulação intracelular de manitol. Neste fungo, o glicerol não é acumulado nas células, quando este poliol é utilizado como fonte única de carbono. Nestas condições, o arabitol continua a ser acumulado em resposta ao aumento da concentração de NaCl no meio, desempenhando, assim, as funções de substância osmorreguladora principal.
Most yeasts and filamentous fungi osmoregulate under water stress by the accumulation of polyols. The intracellular accumulation of polyols in these microorganisms is related to the growth phase and the solute concentration of the growth medium. In the yeasts Debaryomyces hansenii, Candida famata and Pichia farinosa, glycerol is the main compatible solute accumulated in response to water stress. In media with glucose as the sole carbon source, the highest concentration of this polyol is obtained during exponential growth phase. In these yeasts, the substitution of glucose by erythritol, in media with 1 M NaCl, results in the accumulation of large ammounts of this polyol during exponential phase of growth, instaed of glycerol; erythritol replaces intracellular glycerol as compatible solute causing a decrease in the concentration of this late polyol. In the absence of NaCl, all the yeasts accumulate arabitol during late exponential and stationary growth phases. In Geotrichum candidum mannitol is the main polyol detected, in low concentration, during late exponential and stationary growth phases in media without NaCl. In the presence of high NaCl concentration this strain accumulates high levels of arabitol primarily during exponential growth phase. Under these conditions, only traces of mannitol are detected during the late stationary phase. So, in this microorganism, arabitol is the primary compatible solute accumulated in response to the increase of NaCl in the growth medium.
