Internship Report and Monography entitled "Rosacea topical treatment and care: from traditional to new delivery systems"

Abstract

O Mestrado Integrado em Ciências Farmacêuticas da Faculdade de Farmácia da Universidadede Coimbra culmina com a realização do Estágio Curricular em Farmácia Comunitária, sendoeste imprescindível para a formação de profissionais de saúde exímios e competentes noexercício da sua atividade profissional. O presente relatório, estruturado no formato deanálise SWOT, é referente ao meu estágio na Farmácia Central de Fornos Lda. e tem comoobjetivo uma avaliação e reflexão do estágio, tanto a nível interno (Pontos Fortes e PontosFracos) como a nível externo (Ameaças e Oportunidades), que decorreu entre os meses dejaneiro e abril de 2021.O Mestrado Integrado em Ciências Farmacêuticas da Faculdade de Farmácia da Universidadede Coimbra culmina com a realização do Estágio Curricular em Farmácia Comunitária,podendo também realizar Estágio em Indústria Farmacêutica. O presente relatório,estruturado no formato de análise SWOT, é referente ao meu estágio no departamentoQuality Management System dos Laboratórios Basi e tem como objetivo uma avaliação ereflexão do estágio, tanto a nível interno (Pontos Fortes e Pontos Fracos) como a nívelexterno (Ameaças e Oportunidades), que decorreu entre os meses de maio e julho de 2021.A rosácea é uma dermatose inflamatória crónica multifatorial, caracterizada por rubor,eritema persistente, pápulas e pústulas, telangiectasias, e alterações fimatosas, bem comopela presença de prurido, calor, sensação de ardor ou picada, cuja fisiopatologia ainda não étotalmente conhecida. Os tratamentos tópicos convencionais geralmente evidenciam eficácialimitada, devido à propriedade de barreira física da pele que impede a penetração dosingredientes ativos na pele, dificultando a administração adequada dos mesmos, e,consequentemente, os efeitos terapêuticos e/ou cosméticos respetivos. Os novosdesenvolvimentos que se têm verificado a nível da compreensão da fisiopatologia da doença,bem como dos mecanismos subjacentes, sugerem o potencial de novos ingredientes ativoscomo abordagens terapêuticas e cosméticas promissoras para esta dermatose.Adicionalmente, o desenvolvimento de novos sistemas de libertação de fármacos para a pele,particularmente no que concerne ao uso de nanopartículas para o tratamento tópico e parao cuidado da pele com rosácea, tem sido descrito. As particularidades das nanopartículasincidem sobretudo na sua reduzida dimensão nanométrica, que contribui para melhoriasignificativa da capacidade dos ingredientes ativos em atingir os tecidos-alvo. Para além daexposição da fisiopatologia, epidemiologia, diagnóstico e medidas de prevenção, estetrabalho de revisão compila e discute as abordagens tópicas usadas no controle da rosácea,incluindo os cuidados com a pele e cosméticos, e as terapias tópicas, bem como asperspetivas futuras destas estratégias.

The Integrated Master's Degree in Pharmaceutical Sciences at the Faculty of Pharmacy of theUniversity of Coimbra culminates with the completion of the Curricular Internship inCommunity Pharmacy, which is essential for the training of skilled and competent healthprofessionals in the exercise of their professional activity. This report, structured in theSWOT analysis format, refers to my internship at the Central Pharmacy of Fornos Lda. andaims to assess and reflect on the internship, both internally (strengths and weaknesses) andexternally (threats and opportunities), which took place between January and April 2021.The Integrated Master's Degree in Pharmaceutical Sciences at the Faculty of Pharmacy of theUniversity of Coimbra culminates with the completion of the Curricular Internship inCommunity Pharmacy, and may also carry out an Internship in the Pharmaceutical Industry.This report, structured in the SWOT analysis format, refers to my internship in the QualityManagement System department of Basi Laboratories and aims to assess and reflect on theinternship, both internally (Strengths and Weaknesses) and externally (Threats andOpportunities), which took place between May and July 2021.Rosacea is a multifactorial chronic inflammatory dermatosis, characterized by flushing,nontransient erythema, papules and pustules, telangiectasia, phymatous alterations,accompanied by itching, burning or stinging, whose pathophysiology is not yet fullyunderstood. Conventional topical treatments usually evidence limited efficacy, attending tothe physical barrier property of the skin that hinders the skin penetration of the activeingredients, thereby hampering a proper drug skin delivery, and the respective therapeuticand/or cosmetic effects. New advances regarding the physiopathological understanding of thedisease, and the underlying mechanisms, suggest the potential of new active ingredients aspromising therapeutic and cosmetic approaches to this dermatosis. Also, the development ofnew drug delivery systems for skin delivery, particularly the potential of nanoparticles for thetopical treatment and care of rosacea has been described. Emphasis has been laid in theirreduced nanometric size, which contributes to a significant improvement in the obtainmentof a targeted skin drug delivery. Besides the exposition of the known pathophysiology,epidemiology, diagnosis, and preventive measures, this review covers the topical approachesused in the control of rosacea, including skin care and cosmetics, and topical therapies, aswell as the future perspectives of these strategies.
