dc.description.abstract | O cancro é hoje um dos maiores desafios da medicina e a busca de novas abordagens terapêuticas para o seu tratamento é uma importante área do esforço científico mundial. O Liver X Receptor tem-se assumido nos últimos anos como um potencial alvo terapêutico para o tratamento de importantes cancros como o da próstata e o da mama que figuram entre os mais frequentes e letais. A ativação destes recetores em modelos celulares e animais de diversas neoplasias quer pelos oxisteróis, seus agonistas naturais, quer por agonistas sintéticos é capaz de interferir na proliferação e sobrevivência de tumores. Os mecanismos implicados envolvem a homeostase do colesterol, a sinalização hormonal, a interferência no ciclo celular e na angiogénese. Apesar dos resultados promissores, os efeitos adversos resultantes da ativação do LXR sugerem que ainda há muito trabalho a desenvolver para que possam ser encarados como alvos terapêuticos válidos e seguros em humanos. | pt |
dc.description.abstract | Cancer is one of the main challenges of today’s medical sciences and the research of novel therapeutic approaches for its treatment is one of the most important areas of the world scientific effort. The liver X receptor rose in the last few years as a potential therapeutic target for the treatment of diverse cancers including prostate and breast cancers which are among the most frequent and most lethal. The activation of these receptors by either their natural ligands, oxysterols, or by synthetic ones has proved to be capable of arresting cancer cell proliferation and survival. The LXR exerts its effect on cancer cells through diverse mechanisms, which include regulation of cholesterol metabolism, hormone signaling, cell cycle interference and disruption of angiogenesis. Despite the promising results observed in recent years, the adverse effects of LXR activation suggest that there still is a long path to trail in order for LXR to become a valid target for the treatment of cancer in humans. | pt |
