Abstract: A nanotecnologia, onde se inserem as NPs, é uma das áreas mais importantes no desenvolvimento da Indústria Farmacêutica.Sendo as NPs pequenos transportadores onde pelo menos uma das dimensões físicas se encontra entre 1-100 nm, estas contribuem para uma melhoria das características biofarmacêuticas dos princípios ativos.Diversos fatores contribuem para a toxicidade de uma NP, de onde se pode destacar, sem dúvida, a dose administrada, mas também o tamanho e área de superfície, as características da superfície, a estabilidade e a via de exposição.Para que se possa avaliar adequadamente o comportamento das NPs, podem ser realizados estudos in vitro, em culturas celulares, ou in vivo, em animais. Numa situação ideal, apenas seriam utilizadas as culturas celulares, por todas as questões éticas e morais associadas ao uso de animais. No entanto, atualmente, esta situação não se verifica, pois ainda não se estabeleceu uma correlação in vivo/in vitro adequada.O comportamento das NPs acima mencionado corresponde às suas características farmacocinéticas, no que diz respeito à Absorção, Metabolização, Distribuição e Eliminação.Se se entender como é que a partícula se comporta dentro do organismo, e em contacto com os vários fluidos, a compreensão dos seus mecanismos de toxicidade é simplificada.Para finalizar, serão apresentados exemplos práticos de toxicidade de medicamentos já comercializados com lipossomas, NPs poliméricas e metálicas e micelas poliméricas. O efeito secundário mais comum é, de facto, as reações de hipersensibilidade, que podem e devemser prevenidas com a utilização de anti-histamínicos.
Nanotechnology, where NPs can be put in, is one of the most important areas in the development of the Pharmaceutical Industry.As NPs are characterized as small transporters in which, at least, one of the physical dimensions is between 1-100 nm, they certainly lead to an improvement in the biopharmaceutical characteristics of the active ingredients.Many factors can contribute the toxicity of a NP. The dose is undoubtedly the most important one, but we can also mention others like the size and the surface area, surface characteristics, stability and route of exposure.To better assess the behaviour of a NP, two types of studies can be implemented – the in vitro studies, with cellular cultures, or the in vivo studies, with animals. Ideally, only cellular cultures should be used, because of the moral and ethics issues associated to the animal use in such studies. However, nowadays, that does not happen, as an adequate in vivo/in vitro correlation has not been established yet.The above-mentioned behaviour of the NPs is no more than their pharmacokinetic characteristics regarding Absorption, Metabolism, Distribution and Elimination. If we comprehend how a NP behaves inside the organism, and in contact with the various biological fluids, the understanding of its toxicity mechanisms is simplified.Finally, several practical examples of the toxicity of already commercialized medicines with lipossomes, polymeric and metallic NPs and polymeric micelles are presented. The most commonly reported adverse effect is hypersensitivity reactions, which can and shouldbe prevented with the use of antihistamines.
