O potencial de alguns fármacos naturais para o tratamento de doenças neurodegenerativas

Abstract

As doenças neurodegenerativas são um problema que afeta milhões de pessoas no mundo inteiro e cuja prevalência tem vindo a crescer. Há vários fatores que desencadeiam as alterações nas células nervosas, nomeadamente fatores genéticos e fatores extrínsecos, em particular o stress oxidativo. Com o aumento da idade aumenta também a prevalência destas doenças. O excesso de espécies reativas de oxigénio e a perda da capacidade do organismo para limitar e contrariar os seus efeitos, leva a um desequilíbrio que pode culminar com a destruição irreversível das células. As células nervosas não têm a capacidade de se autorrenovarem e, portanto, se sofrerem alguma alteração ou até mesmo se morrerem não há forma de reverter o processo ou de as substituir, o que leva ao aparecimento das doenças neurodegenerativas. Dado não haver tratamento que reverta completamente estas patologias, a prevenção do aparecimento dos fatores desencadeantes e o alívio dos sintomas e consequências são, por hora, as únicas intervenções possíveis para as contrariar. Na presente revisão bibliográfica será descrito o desenvolvimento de diferentes doenças neurodegenerativas,abordando as diferentes causas que estão na origem do aparecimento destas doenças tendo como principal foco o stress oxidativo e a explicação das diferentes abordagens terapêuticas face ao mesmo, tendo como base ingredientes ativos de plantas, tais como os ginsenósidos e a gintonina provenientes da raíz do Panax ginseng, os ginkgólidos e o bilobalido da Ginkgo biloba, a curcumina I, II, III da Curcuma longa e o resveratrol proveniente dos frutos vermelhos, uvas e amendoins.

Neurodegenerative diseases are a major health issue that affects millions of people worldwide and has been growing over time. There are several factors that trigger changes in nerve cells, namely genetic factors and extrinsic factors, such as the oxidative stress. With incresing age, the prevalence of these diseases increases. The excess of reactive oxygen species and the loss of the body's ability to defend itself, called antioxidant capacity, leads to an imbalance and consequently to the appearance of oxidative stress, which can lead to the complete destruction of cells. Nerve cells do not have the capacity to self-renew, so if they suffer any change or even die, there is no way to reverse the process or replace them, which leads to the onset of neurodegenerative diseases. Since no treatment is available for these diseases the prevention of the onset of the triggering factors and the relief of the symptoms are, for now, the unique measures to mitigate them. The present literature review will describe the development of different neurodegenerative diseases,addressing the different causes that are the origin of the appearance of these diseases with the main focus being the oxidative stress and the explanation of the different therapeutic approaches to it, based on active plant ingredients, such as ginsenosides and gintonin from the root of Panax ginseng, ginkgolides and bilobalide from Ginkgo biloba, curcumin I,II and III from Curcuma longa and resveratrol from red fruits, grapes and peanuts.
