dc.description.abstract | A doença de Machado-Joseph (DMJ), também conhecida como ataxia espinocerebelosa do tipo 3, é a ataxia hereditária dominante mais comum no mundo. Esta doença neurodegenerativa progressiva é caracterizada por uma expansão anómala do tripleto CAG no gene ATXN3, que codifica uma cadeia expandida de poliglutaminas na respetiva proteína. Esta mutação resulta num ganho de função tóxico na proteína ATXN3 e, eventualmente, na neurodegenerescência de várias regiões do sistema nervoso central. Atualmente não existe nenhuma terapia eficaz para esta doença fatal. No entanto, dado que a DMJ é causada por uma mutação num gene específico, esta doença é um candidato ideal para terapias que têm como alvo o transcrito mutante. Para além disso, e de forma a preservar a expressão e a função da proteína ATXN3 normal, torna-se fundamental discriminar entre o alelo mutante e o alelo normal. Estudos recentes identificaram ferramentas moleculares robustas para o silenciamento de genes, sendo esta uma das estratégias terapêuticas com maior potencial para o tratamento de doenças de poliglutaminas, onde se inclui a DMJ. O objetivo principal do presente trabalho foi o de investigar o potencial terapêutico de ferramentas moleculares silenciadores de genes que têm como alvos polimorfismos genéticos (SNPs) identificados em 70% dos doentes com DMJ. Como tal, testámos essas ferramentas moleculares num modelo celular e num modelo lentiviral da DMJ, com o intuito de investigar a capacidade de silenciamento, tolerabilidade e seletividade para ATXN3 mutante humana. Concluindo, os resultados do nosso estudo indicam que o uso de moléculas silenciadoras que têm como alvo SNPs associados ao alelo mutante, resultam numa redução significativa dos níveis da proteína ATXN3 mutante no sistema nervoso central de murganhos e sugerem que esta terapia atenua a neuropatologia associada à DMJ num modelo lentiviral da DMJ em murganhos. | por |
dc.description.abstract | Machado Joseph disease (MJD), also known as spinocerebellar ataxia type 3, is the most common dominantly inherited ataxia. This progressive neurodegenerative disorder is characterized by a CAG trinucleotide expansion in the ATXN3 gene, which encodes for an expanded polyglutamine tract in the respective protein, resulting in toxic gain-of-function and eventually in neurodegeneration. Currently, no disease-modifying therapies are available for this fatal disorder. Nevertheless, given its monogenic nature, MJD is an ideal candidate for therapies that target specifically the gene transcript. Moreover, in order to preserve wild-type ATXN3 expression and function, discrimination between wild-type and mutant allele is crucial. Recent studies have identified molecular tools as powerful gene silencing strategies and a promising therapeutic approach for several polyglutamine disorders, including MJD. Here, we performed a proof-of-principle study in which we investigated the therapeutic potential of gene silencing tools targeting SNPs present in 70% of MJD patients. For that purpose, we tested SNP-targeting gene silencing molecules in a cellular and in a lentiviral-based mouse model of MJD to assess silencing capacity, tolerability and selectivity to human mutant ATXN3. Altogether, our proof-of-concept study indicates that molecules targeted to different SNPs displayed potent and selective gene silencing capacities and were well tolerated in neuronal cell lines. Moreover, they result in significant reduction of soluble human mutant ATXN3 protein in the mouse brain and suggest that this therapy may ameliorate neuropathological features in a lentiviral-based mouse model of MJD. | eng |
