Abstract: A presente dissertação teve o objectivo de desenvolver um método analítico através do quality-by-design, com posterior pré-validação para quantificação de substância ativa de uma forma farmacêutica sólida oral de libertação imediata de amostras provenientes de ensaios de dissolução. O método analítico foi desenvolvido em Cromatografia Líquida de Alta Eficiência (HPLC-UV), e foi feita uma abordagem QbD de forma a desenvolver um método mais robusto. Este estudo recorre a modelos estatísticos e a desenho de experiências (DoE). Foram estudadas as diferentes variáveis que afretam o método simultaneamente, e as respectivas respostas, de forma a perceber as suas iterações.O método desenvolvido pretende analisar a substância ativa presente em amostras provenientes de ensaios de dissolução. Este tipo de ensaios é muito utilizado pela indústria farmacêutica, não só para o desenvolvimento de novos medicamentos, mas também em análises de rotina para controle de medicamentos.Os parâmetros pré-validados e foram os seguintes: selectividade, linearidade e gama de trabalho, exactidão, recuperação, repetibilidade, precisão do sistema, estabilidade das soluções e filtração,tendo todos cumprido as respectivas especificações.Este trabalho irá acompanhar o desenvolvimento da formulação do projecto Blue071 através da avaliação e distinção dos diferentes perfis de dissolução, de forma a fazer a diferenciação entre formulações feitas nas condições pretendidas e formulações com alterações nas variáveis mais críticas.
The purpose of the present thesis was to develop, with the Quality by Design approach, and pre-validate an analytical method for quantifying a drug substance in oral solid pharmaceutical form of immediate release from dissolution tests samples.The analytical method was developed in High Performance Liquid Chromatography (HPLC-UV), and a QbD approach was developed in order to develop a robust method. This study applies statistical models and design of experiences (DoE). There were studied different variables that affect the method simultaneously, and the respective responses, in order to understand their interactions. The developed method intends to analyze and quantify the drug substance present in samples from dissolution tests. This type of testing is widely used by the pharmaceutical industry, not only for the development of new drugs, but also for routine analyzes for drug control. When developing a new drug, it is necessary to develop methods to characterize and quantify it in parallel, so that the method can be recognized by the competent authorities so that they can be used in routine tests or in bioequivalence tests of that medicinal product. The pre-validated parameters,within all the specifications, were as follows: selectivity, linearity and working range, accuracy, recovery, repeatability, system precision, solutions stability and filtration.This work will be used for development the formulation of the Blue071 project by evaluating and distinguishing different dissolution profiles in order to distinguish between formulations made under the desired conditions and formulations with changes in the most critical variables.
