Abstract: A monografia apresenta o trabalho executado na Universidade de Liège, Bélgica, aquando do estágio no âmbito do programa ERASMUS +. Este consistiu na continuação da investigação de compostos com atividade anti-plasmódica, nomeadamente, o composto Poupartone B extraído da planta Poupartia borbonica, endémica às Ilhas Mascarenhas. Este composto é o segundo da família de compostos extraídos desta planta a ser estudado quanto à sua possível aplicação no tratamento da malária cerebral. O trabalho experimental englobou diversos passos da investigação: extração, isolamento e purificação dos compostos, construção de lipossomas, validação e avaliação da estabilidade da formulação, quantificação da heparina, estudos de toxicidade num modelo com zebrafish e testes de eficácia in vitro em estirpes de P. falciparum. Durante a investigação novos compostos foram descobertos e investigados em simultâneo. A validação e estabilidade foram comprovadas e a aplicabilidade da formulação averiguada por meio dos ensaios referidos. Os novos compostos não revelaram qualquer eficácia, no entanto a formulação com Poupartone B proporcionou uma redução significativa da toxicidade comparativamente à utilização do composto isolado, bem como o aumento da eficácia do mesmo no parasita in vitro, viabilizando o seguimento da investigação para ensaios in vivo. Os relatórios de estágio estão sob a forma de Análise SWOT. Esta consiste numa análise integrada dos fatores internos e externos aos estágios, e de como estes foram percecionados. Desta forma, as experiências são descritas com base numa correlação entre forças, fraquezas, oportunidades e ameaças. O estágio nos Serviços Farmacêuticos da Unidade Local de Saúde de Castelo Branco permitiu o contato com o hospital, os circuitos do medicamento e a especialidade farmacêutica. O estágio na Farmácia Nuno Álvares possibilitou uma experiência de farmácia comunitária. Proporcionou, igualmente, aprendizagens quanto ao papel do farmacêutico, o contato com os utentes, e o modo de funcionamento de uma farmácia.
This report presents the work carried out at the University of Liège, in Belgium, through the ERASMUS + program. It consisted of continuing the research on compounds with anti-plasmodial activity, namely, the Poupartone B compound extracted from Poupartia borbonica, a plant endemic to the Mascarene Islands. This compound is the second of its family of compounds to be studied for a possible application in the treatment of cerebral malaria. The experimental work consisted of several research stages: extraction, isolation and purification of the compounds, construction of liposomes, validation and evaluation of the formula’s stability, heparin quantification, toxicity studies in a zebrafish model and efficacy tests in in vitro P. falciparum strains. In the process, new compounds were discovered and simultaneously investigated. The validation and stability were successfully demonstrated and the applicability of the formulation was assessed. The newly discovered compounds proved to have no action against the parasite, however, the formulation with Poupartone B managed to significantly reduce the toxicity when compared to the compound alone, as well as augment the efficacy against the parasite in vitro, allowing the follow up of the investigation to in vivo tests. The internship’s reports are written in SWOT analysis. This form of analysis consists of an integrated analysis of the external and internal factors that influence the internships and how they were perceived. This way, the experiences are described through means of a correlation between strengths, weaknesses, opportunities and threats. The internship at the Pharmaceutical Services of the Castelo Branco Local Health Unit allowed contacting with the hospital, the medicines’ circuits and the pharmaceutical specialty. The internship at the Nuno Álvares Pharmacy allowed experiencing in community pharmacy. It enabled the learning process about the pharmacist’s role, how to contact with patients and how a pharmacy works.
