dc.description.abstract | As micotoxinas são metabolitos secundários produzidos por fungos que ocorrem de forma natural nos vários produtos agrícolas. A contaminação de alimentos por estes fungos ocorre em condições ambientais durante o crescimento das culturas, nas colheitas inadequadas, assim como, durante o transporte e mais frequentemente no armazenamento.A ocratoxina A (OTA) é uma micotoxina produzida por alguns de fungos, tais como, Penicillium verrucosum, Aspergillus ochraceus e o Aspergillus niger. A OTA é conhecida principalmente pela sua nefrotoxicidade, apesar de possuir outras propriedades tóxicas. É também um composto tóxico cumulativo pela sua facilidade de absorção pelo aparelho digestivo mas de excreção lenta, através da via urinária. Está ainda classificada pela IARC como um possível carcinogénico humano, grupo 2B.De entre as várias ocratoxinas, a OTA é considerada a mais tóxica, sendo a mais relevante para a segurança alimentar. Por este motivo, considerou-se essencial detetar e quantificar a concentração de OTA em cerveja consumida em Portugal.A linearidade da OTA avaliada foi adequada, com valores em fase móvel (2,5 – 25 ng/mL) de R2=0,998, e na matriz (0,25 – 2,5 µg/L) de R2=0,9985. O valor do efeito matriz obtido foi de 100,05%. As recuperações variaram entre 81% e 86%. O LOD e o LOQ foram de 0,14 e 0,43 µg/L, respetivamente.Neste estudo foi determinada a concentração de OTA em 33 amostras de cerveja, em que 9 amostras (27,27%) estavam contaminadas, com teores que variaram entre 0,174 e 11,25 µg/L, sendo o teor médio de 3,14 µg/L. Duas das amostras de cerveja de fabrico caseiro apresentaram concentrações consideráveis, com teores de 9,21 µg/L e 11,25 µg/L. dc.description.abstract | Mycotoxins are secondary metabolites produced by fungi which occur naturally in several agricultural products. Food contamination by these fungi occurs under environmental conditions during crop growth, inadequate harvests, and also during transportation and storage, which is more often. Ochratoxin A (OTA) is a mycotoxin which is produced by some species of fungi, such as Penicillium verrucosum, Aspergillus ochraceus and Aspergillus niger. OTA is mainly known for its nephrotoxicity, although it possesses other toxic properties. It is also a cumulative toxic compound because it is easily absorbed by the digestive system and is slowly excreted through the urinary system. OTA was classified by IARC as a possible human carcinogen, group 2B.Among the several ochratoxins, OTA is considered to be the most toxic, and it is also the most relevant to food safety. For this reason, it was deemed essential to detect and quantify the concentration of OTA in the beer consumed in Portugal. The OTA linearities were adequate, with values of R2 in mobile-phase (2.5 – 25 ng/mL) and in the matrix (0.25 – 2.5 µg/L) of 0.998 and 0.9985, respectively. The value of the matrix effect obtained was 100.05%. Recoveries ranged between 81% and 86%. LOD and LOQ were of 0.14 and 0.43 µg/L, respectively.The OTA concentration was determined in 33 beer samples, 9 of which (27.27%) were contaminated, with levels ranging between 0.174 and 11.25 µg/L, and an average level of 3.14 µg/L.
