Abstract: Compreender como os diferentes membros do Sistema imunitário trabalham em conjunto para nos proteger contra a doença tem um forte impacto na saúde pública. Células Linfoides Inatas (ILCs) são o mais recente componente do sistema imunitário e a sua importância tem sido altamente reconhecida, como critica na defesa do hospedeiro, em moldar o microbiota e na reparação tecidual.Estas células foram apenas estudadas e descritas em humano e rato. O objetivo deste trabalho será tentar possível a existência e o papel dos diferentes subtipos de ILCs e também a conservação da função da citoquina IL-22 em peixe-zebra. IL-22 é uma importante citoquina recentemente identificada em mamíferos e os limitados dados sugerem um papel protetivo da mucosa. Contudo, a sua função e as células produtoras são informações ainda desconhecidas em peixe-zebra. O peixe-zebra (Danio rerio) é um vertebrado geneticamente manipulável de acessibilidade ótica notável o que faz dele um excelente modelo para responder às questões propostas para desenvolver este trabalho. Para identificar os subtipos de ILCs, populações celulares serão isoladas usando citometria de fluxo e o seu perfil transcricional será avaliado em massa e ao nível de single-cell. Para estudar a função da citoquina IL-22, mutantes para esta citoquina irão ser gerados. Juntamento com a linha transgénica repórter para il22 a identificação de células produzindo IL-22 será “desafiada” com agentes patogénicos ou através de estímulo inflamatório e então estudada.
Understanding how the different members of the immune system work together to protect us against disease has a strong impact in public health. Innate lymphoid cells (ILCs) are the most recently discovered component of the immune system and their importance has been highly recognised, as critical in host defence, in shaping the microbiota and in tissue repair. ILCs have been only studied in mice and humans but so far there are no reports of ILCs in other species. This project will address the existence and role of the different subsets of ILCs and the conservation of IL­22 function in zebrafish. The IL-22 is an important cytokine recently identified in mammals and the limited data suggests a protective role in the mucosae. However, the function of IL-22 and the identity of the cells producing this cytokine remain unexplored in Zebrafish. The Zebrafish (Danio Rerio) is a genetically tractable vertebrate with remarkable optical accessibility which makes it a great model to address these questions. To identify ILC subsets, cell populations, enriched in putative ILCs will be isolated by flow cytometry and their transcriptome profile will be assessed in bulk and at the single-cell level, by qPCR. To study IL­22 function, mutants for this cytokine and its receptor will be generated. Together with a transgenic il22 reporter line allowing the identification of IL­22 producing cells, they will be challenged with pathogens or inflammatory stimuli, and then studied by various approaches.
