estruturais, que possibilitaram criar réplicas do envelope viral, as quais permitiram a
realização de estudos in vitro para avaliar a capacidade e a forma de indução da produção de
AC capazes de travar a infeção pelo vírus. Passaram 30 anos sem se conseguir uma vacina
capaz de neutralizar o vírus, impedindo a propagação da infeção. Numerosos estudos foram
iniciados mas poucos foram concluídos, e, no universo dos que chegaram ao fim, apenas
alguns obtiveram resultados positivos, sendo RV144 o ensaio com melhores resultados até
hoje, com 31, 2% de eficácia. Nos ensaios VAX004 e VAX003, os primeiros ensaios de Fase
III realizados em Humanos, a vacina em estudo não demonstrou qualquer eficácia no
entanto, o ensaio foi considerado importante pois permitiu perceber o que não resulta
como vacina, sendo que depois a seu AG foi utilizado como reforço no ensaio RV144. Muitos
outros estudos foram realizados depois deste, alguns chegando mesmo a utilizar R144 como
base, tal como HVTN097, outros utilizando antigénios e regimes de vacinação diferentes
(HVTN092), mas sempre com o mesmo objectivo: obter uma vacina o mais rapidamente
possível para o controlo desta doença. Since it was discovered, HIV has been the subject of many investigations. The structural analyzes, which permitted to create reproductions of the viral envelope, allowing in vitro studies to assess the capacity of induction, and shape, of Antibody (AB) that are able to halt the infection. However, 30 years have passed without achieving a vaccine capable of neutralizing the virus and preventing the spread of the infection. Many studies have been conducted but few have been completed, and even those who come to an end, only a few of them had positive results, being the RV144 trial the one with best results to date, with 31.2% efficacy. VAX004 and VAX003, the first Phase III trials conducted in Humans, and although the results have not shown any effectiveness, the realization that it was ineffective as a vaccine has guaranteed the study a place of high importance, providing the AG as boost in RV144 . Many other studies have been carried out after this, some using R144 as base, such as HVTN100, and other using different antigens and vaccine regimens (HVTN092), with the same objective: obtain a vaccine as quickly as possible to control this disease. |
