Title: Lesão celular e controlo de qualidade proteico : reparação versus degradação

Abstract: O primeiro objectivo deste trabalho consistiu em determinar qual a via proteolítica envolvida na remoção das proteínas modificadas pelo 4-hidroxinonenal (HNE) nas células. Os resultados obtidos mostraram que a a-cristalina modificada pelo HNE é monoubiquitinilada, embora não se tenha observado alterações significativas na susceptibilidade para a degradação pelo proteossoma em comparação com a a-cristalina nativa. A microinjecção de a-cristalina em células epiteliais do cristalino em cultura mostrou que a a-cristalina modificada pelo HNE é mais susceptível à degradação que a a-cristalina nativa. Em células tratadas previamente com HNE, a depleção de ATP ou a presença de cloroquina (inibidor do lisossoma) estabiliza as proteínas modificadas pelo HNE. Contudo, inibidores específicos do proteassoma não inibem a remoção das proteínas modificadas pelo HNE em células tratadas previamente com HNE. Com base nos resultados neste estudo, conclui-se que as proteínas modificadas pelo HNE são degradadas pelo lisossoma de um modo dependente da ubiquitina. Neste trabalho foi dada especial atenção ao sistema de controlo de qualidade das proteínas, fundamental para a manutenção da homeostase e sobrevivência celular. A luciferase desnaturada pelo calor é preferencialmente ubiquitinilada e degradada de modo dependente de Hsp90 e CHIP. No entanto, a adição de Hsp70 inibe a degradação das proteínas desnaturadas. Por outro lado, a inibição da ubiquitinilação ou da proteólise aumenta a renaturação da luciferase. Os resultados obtidos sugerem que quer a via proteolítica dependente da ubiquitina e proteossoma, quer os chaperones moleculares reconhecem os mesmos motivos nas proteínas lesadas. Por outro lado, a via proteolítica da ubiquitina e proteossoma e os chaperones actuam de um modo competitivo. A actividade desubiquitinilante pode constituir uma nova oportunidade das proteínas serem reparadas.
The first objective of the work was to determine HNE-modified proteins are removed from cells. The data showed that _B-crystallin modified by HNE was ubiquitinated at a faster rate than that of native _B-crystallin in a cell-free system. When delivered into cultured lens epithelial cells, HNE-modified _B-crystallin was degraded at a faster rate than that of unmodified _B-crystallin. Inhibition of the lysosomal activity stabilized HNE-modified _B-crystallin, but inhibition of the proteasome activity alone had little effect. Depletion of ATP or the presence of chloroquine, a lysosome inhibitor, stabilized HNE-modified proteins. However, proteasome-specific inhibitors could not stabilize HNE-modified proteins in the cells. The enrichment of HNE-modified proteins in the fraction of ubiquitin conjugates suggests that HNE-modified proteins are preferentially ubiquitinated. Taken together, these findings show that HNE-modified proteins are degraded via a novel ubiquitin and lysosomal-dependent but proteasome-independent pathway. In this work special attention was given to the protein quality control system because this system plays a fundamental role in normal cellular function. The data shown that thermally denatured luciferase is preferentially ubiquitinated and degraded by the ubiquitin-proteasome pathway (UPP) in Hsp90 and CHIP dependent manner. Addition of Hsp70 inhibits degradation of denatured protein. On the other hand, the inhibition of the ubiquitination or proteasome activity enhanced renaturation activity. This work demonstrates for the first time that the UPP and molecular chaperones work in a competitive manner and that the fates of denatured proteins are determined by the relative activities of the UPP and molecular chaperones. These data suggest that the UPP recognizes the same features of denatured proteins that are recognized by molecular chaperones and that Hsp90 was used by the UPP in the recognition process. Isopeptidase activity is also a factor in determining the fate of denatured proteins, prevent protein degradation and play a role in the refolding process.
