Abstract: Os medicamentos podem ver a sua eficácia e segurança comprometida como resultado da presença de diferentes fases sólidas do API, durante o processo de fabrico e de armazenamento. Isto implica a necessidade de investigação do polimorfismo e das suas consequências na qualidade do produto acabado. Por outro lado, a pesquisa de novas formas sólidas constitui, por si só, um desafio para a indústria farmacêutica visando encontrar a melhor solução para a otimização de propriedades físico-químicas no contexto de investigação e desenvolvimento. Este projeto tem por objetivo a investigação e a caracterização de novas formas polimórficas de dois ativos farmacêuticos, o salbutamol, usado para o tratamento da asma e o iguratimode, útil para o tratamento da artrite reumatoide. Ambos mostraram uma forte dinâmica comercial no mercado global. Assim, o screening de novas formas sólidas é realizado recorrendo à cristalização em solução. O estudo dos sólidos obtidos é feito por espectroscopia de infravermelho, por métodos de análise térmica (calorimetria diferencial de varrimento e termomicroscopia) e, ainda, por difração de raios-X de pó. O estudo preliminar dos sólidos obtidos por cristalização, permitiu-nos identificar algumas formas sólidas, novas, do iguratimode, sendo uma delas um solvato. No caso do salbutamol foi registada pela primeira vez evidência de uma forma polimórfica.
The medicaments may see their efficacy and safety compromised as a result of the presence of different solid phases of the API, during the process of manufacture and storage. This implies the need for research of polymorphism and of its effect on the quality of the finished product. On the other hand, the search for new solid forms is, in itself, a challenge to the pharmaceutical industry, aimed at finding the best solution for the optimization of physicochemical properties in the context of research and development. This project aims at investigating and characterizing new polymorphic forms of two active pharmaceutical ingredients, salbutamol, used to treat asthma, and iguratimode, useful for the treatment of rheumatoid arthritis. Both showed strong commercial dynamics in the global market. Thus, the screening of new solid forms is performed using crystallization from solutions. The study of the obtained solids is made by infrared spectroscopy, thermal analysis methods (differential scanning calorimetry and termomicroscopy) and also by X-ray powder diffraction. The preliminary study of the solid samples obtained by crystallization allowed us to identify some new solid forms of iguratimode, one of then a solvate. In the case of salbutamol evidence of a polymorphic form is registered for the first time.
