Introduction: The muscle contraction activation depends on a stimulus that arises from the transmission of a nervous impulse known as action potential. The action potential results in the inversion of the electrochemical gradient throughout the neuron creating the electric flow. We acknowledge that the muscle contraction settles in the big fundamentals of electricity. Acupuncture consists in a peripheral sensitive and motor nerve stimulation technique that activates many auto-regulatory mechanisms in the human body. Objectives: This experimental study aims to harvest the action potential data, in a surface study, through evaluation of the electrical activity in the practice of dry puncture, in the masseter. Methods: There were selected 30 volunteers. The protocol of each session consisted of the puncture of 3 holes: 3 cm ahead of the 05 San Jiao hole; top region of the masseter muscle – channel 0 and bottom region of the masseter muscle – channel 1, with combined electrode-needle systems to record the electrical potentials, during 5 minutes. Results: In the channels 0 and 1, the average power density (μV2/Hz) for all volunteers was 9,22 ± 1,45 e 9,46 ± 1,77, respectively. On both channels, the average voltage root mean square (μV) was 37,31 ± 280,53 e 55,67 ± 377,07, respectively. For channel 0, the voltage’s average value for the first minute was 7098,29 ± 998,47, 7684,34 ± 1052,05 for the second minute, 7098,29 ± 998,47 for the third minute, 5840,94 ± 828,15 for the fourth minute and 4853,85 ± 626,56 for the fifth minute. For channel 1, the voltage’s average value for the first minute was -9204,35 ± 2942,04, -7246,53 ± 1042,39 for the second minute, -5691,07 ± 1095,92 for the third minute, -7685,49 ± 836,32 for the fourth minute and -10867,78 ± 650,55 for the fifth minute. The results showed statistically significant values between channel 0’s voltage, in the first and second minutes, and the emotional state, p = 0,01 and p = 0,04, respectively. For channel 1, statistically significant values where found between this channel’s voltage values, in every minute, and the emotional state: p = 0,02, p = 0,05, p = 0,03, p = 0,03 e p = 0,03 and the fatigue between the second and fifth minutes: p = 0,04, p = 0,04, p = 0,05 e p = 0,04, respectively. The value of channel 0’s power density and systemic disease was p = 0,03. Discussion: The channel 0 potentials decreased in every subsequent minute, whereas in channel 1, is first noticed a decrease, followed by an increase in the action potential. These results show a statistically significant relation between emotional state and the variation of the action potentials. Conclusions: This study allowed an in-depth analysis of the physicochemical reactions that occur in our organism during acupuncture. Keywords: Action potential, electricity, acupuncture, masticatory muscles.
