O
exercício
físico
tende
a
aumentar
as
células
endoteliais
progenitoras
(EPCs)
na
circulação
sanguínea.
Estas
células,
constituídas
por
varias
populações,
promovem
o
aparecimento
de
novos
vasos,
quer
por
angiogénese,
quer
por
neovascularização.
Assim,
podemos
assumir
que
a
vascularização
aumenta
com
a
prática
de
exercício
físico.
Estudos
recentes
mostram
que
inibidores
de
COX-‐2
são
capazes
de
inibir
a
actividade
de
EPCs
e,
por
tanto,
originar
uma
diminuição
do
aparecimento
e/ou
ramificação
de
novos
vasos.
Este
estudo
pretende
provar
que
o
ibuprofeno,
como
inibidor
da
via
das
ciclooxigenases,
consegue
diminuir
a
vascularização
no
músculo
solear.
Metodos:
Quatro

De
seguida
foram
retiradas
amostras

As

Resultados:
Verificámos
que
com
exercício
a
massa
corporal
aumentou
a
cima
de
todos
outros
grupos,
enquanto
que
o
grupo
de
exercício
com
administração
de
ibuprofeno
apresentou
valores
bastante
abaixo
dos
outros
grupos.
Pode
observar-‐se
também,
que,
não
só
houve
um
aumento

e
uma
diminuição
no
caso
do

de
ibuprofeno.
O
teste
de
performance
efectuado
após
o
treino
mostrou
menor
eficácia
no
grupo
que
consumiu
ibuprofeno.
As
análises
ao

revelaram
que
nos
grupos
de
exercício
existiu
um
aumento
de
VEGF
e
diminuição
dos
valores
de
células

em
circulação,
o
que
se
reflectiu
num
aumento
da
razão

Já
no
grupo
de
exercício
com
administração
de
ibuprofeno,
existiu
uma
diminuição
de
VEGF
e
um
aumento
do
nível
de

CD133+
e

o
que
se
correlacionou
com
a
diminuição
da
razão

quando
comparado
com
o
grupo
de
exercício.
Conclusão:
Não
só
o
exercício
físico
revela
uma
maior
activação
de
EPCs
na
corrente
sanguínea,
como
o
ibuprofeno
parece
ter
a
capacidade

inibir
a

dessas

impedindo
que
as

obrigando-‐as

Acresce
o

treino. Background: Physical Exercise tends to increase endothelial progenitor cells (EPCs) in the bloodstream. These cells, comprised of several populations, promote the development of new blood vessels, either through angiogenesis or by neovascularization. Thus, we can assume that the vasculature increases with the practice of exercise. Recent studies show that COX-‐2 inhibitors are able to inhibit the activity of EPC and, therefore, cause a decrease in the onset and/or branching of new vessels. This study aims to prove that ibuprofen, as an inhibitor of cyclooxygenase pathway, can decrease vascularity in the gastrocnemius and soleus muscles. Methods: Four groups of Wistar rats were oblied to participate in continued aerobic training, on treadmill, over eight weeks. Then blood samples and soleus muscles were collected. The tissue samples were subjected to molecular and histological analysis while the blood samples were used to study of endothelial progenitor cells (EPCs) and VEGF quantification. After, they were subjected to flow cytometry using CD34, CD133, CD146 and KDR markers. VEGF concentrations were determined through ELISA technic. The tissue samples lead to the quantification of muscle fibers as well as the capillary/fibre racio. Results: We confirm that the exercise group was the one with the most body mass increase, above all other groups. It was also clear that the exercise group with administration of ibuprofen showed values well below the other groups. It is also to be noted that, not only there has been an increase of food eaten by the subjects with exercise and a reduction related to the exercise group with administration of ibuprofen. The performance test carried out after the training showed low efficacy in the group that consumed ibuprofen. The analyzes revealed that the blood plasma belonging to the exercise group had an increase of VEGF molecule and decreased values of KDR+, CD146+ cells in circulation, which was reflected in an increase of the fiber/capillary racio. In the group of exercise with administration of ibuprofen, there was an inverse scenario. We saw a decrease in VEGF and increased levels of KDR+, CD146+, CD133+ and CD34+ cells in circulation, correlating to a decrease in the capillary/fiber ratio when compared to the group of only exercise bouts. Conclusions: Not only does exercise reveals a greater activation of EPCs into the bloodstream, but also ibuprofen appears to have the ability to inhibit the activity of these vascular cells, and thus, preventing differentiation into mature endothelial cells (EC), forcing EPCs to remain in the bloodstream for extended periods of time. Moreover, the is a decrease of the vascularization that is correlated with a lesser amount of fibres type I and a decrease in performance after training |
