induzir a coagulação do sangue. Um dos grandes desafios da ciência dos
biomateriais é o desenvolvimento de materiais que minimizem ou anulem a
coagulação sanguínea, melhorando a hemocompatibilidade de dispositivos
médicos. No presente estudo, foram preparadas monocamadas autoestruturadas
(SAMs) sobre ouro de 1-undecanotiol (UDT) e ácido 11-mercapto-
1-undecanoico (MUA), que possuem diferentes grupos químicos à superfície
(UDT: grupos metilo; MUA: grupos carboxilo), de forma avaliar o efeito da
hidrofilicidade/hidrofobicidade e da carga elétrica das superfícies sobre alguns
aspetos da coagulação do sangue: a coagulação do plasma sanguíneo, a adesão
de plaquetas e a adsorção de proteínas. Verificou-se que as proteínas do
plasma adsorviam fortemente à SAM de UDT, hidrofóbica e apolar. Nesta
superfície, também ocorreu uma elevada adesão de plaquetas, explicável pela
interligação entre a adsorção de proteínas e a adesão celular. A SAM de UDT
induziu uma menor ativação das vias intrínseca e extrínseca da coagulação
quando comparada com a SAM de MUA, ou seja, a superfície hidrofílica e com
carga elétrica negativa, ativou mais fortemente a coagulação do plasma
sanguíneo, pelas vias intrínseca e extrínseca. Relativamente à adesão de
plaquetas nas superfícies, verificou-se que na SAM com características
hidrofóbicas e apolar, as plaquetas aderiam em maior quantidade do que à
SAM hidrofílica e com carga elétrica negativa. Nos estudos de adsorção de
proteínas do plasma à superfície das diferentes SAMs, verificou-se que, em
ambas as SAMs, havia adsorção de proteínas do plasma havendo, no entanto,
uma maior adsorção de proteínas à superfície da SAM com características
hidrofóbicas e apolar. The contact between blood and the surface of certain materials can induce blood clotting. One of the great challenges of the science of biomaterials is the development of materials that minimize or eliminate blood clotting, improving the hemocompatibility of medical devices. In this study, selfassembled monolayers of alkanethiols on gold (SAMs) were prepared with 1- undecanotiol (UDT) and with 11-mercapto-1-undecanoic acid (MUA), two molecules which have different chemical groups exposed at the surface (UDT: methyl groups; MUA: carboxyl groups) and used to evaluate the effect of the hydrophilicity/hydrophobicity and the electric charge of the surfaces on some aspects of blood coagulation, such as blood plasma clotting, platelet adhesion and protein adsorption. It was found that plasma proteins strongly adsorb to the hydrophobic and nonpolar UDT SAM. On this surface, there is also a high adhesion of platelets, which can be explained by the interconnection between protein adsorption and cell adhesion. The UDT SAM also induced a lower activation of the intrinsic and extrinsic pathways of coagulation when compared to the hydrophilic and negatively charged MUA SAM, which strongly activates coagulation of blood plasma through the intrinsic and extrinsic pathways. With regard to platelet adhesion on surfaces, platelets adhere in greater quantity to the SAM with hydrophobic and nonpolar characteristics than to the SAM with hydrophilic character and negative electric charge. In the plasma protein adsorption studies, there is plasma protein adsorption on both SAMs, however, there is a greater adsorption of plasma proteins on the surface of the SAM with hydrophobic characteristics and nonpolar. |
