Abstract: Introduction: Hyperglycemia and tissue ischemia are two main features of most of diabetic complications. It has been suggested that high glucose impairs the cell and tissue adaptation to low oxygen, leading to cell death and tissue dysfunction. The loss of cell response to hypoxia, observed in most of diabetic tissues, appears to result from a destabilization of the transcription factor HIF-1 (Hypoxia-Inducible Factor-1) induced by high levels of glucose or methylglyoxal (MGO), a well-known by-product of glycolysis. However, the molecular mechanisms involved in this process remain elusive. Objectives: This study aims to clarify the molecular mechanisms that may be involved in the destabilization of HIF-1α in the presence of high glucose or high levels of MGO. Methodology: A human cell line from retinal pigment epithelium (ARPE-19) was used in this study to investigate the role of high glucose or high levels of MGO in the regulation of HIF-1α. Immunocytochemistry, western blot and immunoprecipitation assays were used to determine the cellular distribution, stability and ubiquitination of HIF-1α, respectively. We further used short-hairpin RNAs (shRNAs) against CHIP to test the function of this ubiquitin ligase in the destabilization of HIF-1α in the presence of MGO or high glucose. Results: Data shows that MGO and high glucose induce downregulation of HIF-1α in ARPE-19 cells, under normoxia and hypoxia conditions. Additionally, results indicate that MGO decreases the accumulation of HIF-1α into the nucleus, under hypoxia, and decreases the expression of VEGF-A, a well-known target gene of HIF-1. These data suggests that MGO decreases the transcriptional activity of HIF-1, under hypoxia conditions. Data further shows that CHIP, which is an ubiquitin ligase involved in protein quality control mechanisms, has an important role in the ubiquitination and destabilization of HIF-1α in the presence of increased levels of MGO or high glucose, under hypoxia conditions. 2 Conclusions: This work suggests that the loss of cell response to hypoxia in diabetes might be associated with increased degradation and decreased stability of the transcription factor HIF-1, due to the activation of a mechanism, which involves ubiquitination of HIF-1α by CHIP, induced by high levels of MGO or high glucose.
Introdução: A hiperglicemia e a isquémia tecidular são duas características importantes de muitas das complicações da diabetes. Tem vindo a ser sugerido que níveis elevados de glicose comprometem a adaptação celular a níveis reduzidos de oxigénio, conduzindo a morte celular e disfunção tecidular. Essa perda de reposta celular à hipoxia, característica da diabetes, parece resultar de uma destabilização do factor de transcrição Hypoxia Inducible Factor-1 (HIF-1) induzida por níveis elevados de metilglioxal (MGO), um produto secundário da glicólise, ou alta glicose. No entanto, os mecanismos moleculares envolvidos neste processo continuam ainda pouco claros. Objectivos: Este trabalho tem como objectivo esclarecer alguns dos mecanismos moleculares que podem estar na origem da destabilização do HIF-1α na presença de alta glicose ou níveis elevados de MGO. Metodologia: Neste trabalho foi usada uma linha celular humana de epitélio pigmentado da retina (ARPE-19) para testar os efeitos da alta glicose ou níveis elevados de MGO na expressão de HIF-1α. Foram usadas técnicas de imunocitoquímica, western-blot e imunoprecipitação para determinar a localização celular, estabilização e ubiquitinação do HIF-1α, respectivamente. Foram ainda testados short-hairpin RNAs (shRNAs) contra a CHIP, para testar a função desta ligase de ubiquitina na destabilização do HIF-1α na presença de MGO ou alta glicose. Resultados: Os resultados obtidos mostram que o MGO e concentrações elevadas de glicose diminuem os níveis de HIF-1α, em células ARPE-19, em condições de normoxia e hipoxia. Verificou-se ainda que o MGO diminui a acumulação de HIF-1α no núcleo e diminui a expressão de VEGF-A, um gene alvo do HIF-1, o que poderá indicar uma diminuição da actividade de transcrição deste factor. Os resultados indicam ainda que a CHIP, uma ligase de 4 ubiquitina envolvida no controlo de qualidade proteica, tem uma função importante na ubiquitinação e destabilização do HIF-1α na presença de níveis elevados de MGO ou glicose em condições de hipoxia. Conclusões: Este trabalho sugere que a perda de resposta celular à hipoxia na diabetes pode estar associada a um aumento da degradação e diminuição da estabilidade do factor de transcrição HIF-1, através de um mecanismo que envolve a sua ubiquitinação, pela ligase de ubiquitina CHIP, induzida por níveis elevados de MGO ou alta glicose.
