Title: O impacto da restrição calórica no envelhecimento

Abstract: A dieta de restrição calórica tem potencial para contrariar o aparecimento de patologias associadas à idade, reduzir o ritmo de envelhecimento, e aumentar a longevidade, intervindo tanto no envelhecimento primário como no secundário. Os seus efeitos vão muito para além da diminuição do peso corporal. Praticamente todos os fatores de risco da doença aterosclerótica são melhorados, o que juntando ao decréscimo dos mediadores inflamatórios e à diminuição de fibrose, vai preservar a integridade da função vascular durante o envelhecimento. A RC revelou diminuir o risco de diabetes mellitus tipo 2, e aumentar a adiponetina; os seus efeitos na insulina são complexos mas apontam no sentido de aumentar a sobrevivência. A RC retarda também a deterioração do SNA, tem um papel favorecedor no cérebro, especialmente se este já for envelhecido, e mostra-se capaz de reduzir o aparecimento de neoplasias malignas. Através da modulação do stress oxidativo, a RC afeta amplamente processos inerentes à deterioração do organismo com o tempo, como a peroxidação lipídica, a oxidação de proteínas, o dano do DNA nuclear e mitocondrial, o dano do RNA. Ao estimular a atividade de sirtuínas, a RC consegue estabilizar a transferência de eletrões na CTE, aumentar a biodisponibilidade da eNOS (diminuindo a pressão arterial), modular a transcrição de genes pela desacetilação de histonas, o que implica a compactação da cromatina, e intervir ainda na prevenção da doença de Alzheimer.
The diet of calorie restriction (CR) has the potential to counteract the emergence of ageassociated pathologies, to reduce the rate of aging, and to increase longevity, intervening in both primary and secondary aging. CR effects go far beyond the decrease in body weight. Almost all the risk factors for atherosclerotic disease are improved, which associated to the decrease of inflammatory mediators and of fibrosis will preserve the integrity of the vascular function during aging. CR revealed to decrease the risk of type 2 diabetes mellitus, and increase the adiponectin; CR effects on insulin are complex but they indicate an increase in the survival. RC also delays SNA deterioration, has a protective role in the brain, particularly in the aging brain, and shows capacity to reduce the appearance of cancer. Through the modulation of oxidative stress, RC widely affects processes inherent to the deterioration of the body over time, such as lipid peroxidation, protein oxidation, and damage of nuclear and mitochondrial DNA and RNA. By stimulating the activity of sirtuins, CR can stabilize the electron transfer in the electron transport chain, increase the bioavailability of eNOS (which will decrease blood pressure), modulate genetic transcription by deacetylation of histones, which implies the compaction of chromatin, and it can even intervene in the prevention of Alzheimer's disease.
