Vários estudos efectuados a nível mundial têm associado genes do complexo SNARE à PHDA, particularmente o gene SNAP-25. Contudo, não existe nenhum estudo efectuado relativamente ao gene SNAP-25 na população portuguesa. Assim, no sentido de se investigar uma eventual associação entre o polimorfismo T1069C no gene SNAP-25 e a PHDA na população portuguesa, procedeu-se à recolha de amostras de probandos e seus progenitores (trios). A genotipagem foi efectuada por PCR-RFLP e os dados obtidos foram analisados segundo as estratégias do HRR e TDT e os resultados revelaram uma tendência de associação entre o polimorfismo T1069C do gene SNAP-25 e a PHDA (HRR: c2=3,50; d.f =1; p=0,061; TDT: c2=3,75; d.f =1, p=0,053).
Assim, a estratégia de associação com base em trios e métodos HRR e TDT, que revelaram uma tendência de associação entre o alelo T do polimorfismo T1069C do gene SNAP-25 e a PHDA, parece sugerir que o polimorfismo estudado poderá desempenhar um papel directo na etiologia da PHDA na população portuguesa. Será importante o uso de uma amostra maior, bem como a análise de haplotipos, a fim de esclarecer uma eventual associação entre o gene SNAP-25 e a PHDA na população portuguesa Attention Deficit and Hyperactivity Disorder (ADHD) is a neuropsychiatric disorder that affects 8% to 12% of children worldwide and is associated with increased risk of learning difficulties, alcohol abuse/dependence and antisocial personality disorders. Several studies worldwide have associated the ADHD genes with the SNARE complex, particularly the SNAP-25 gene. However, there is no study carried out for the SNAP-25 gene in the Portuguese population. In order to investigate a possible association between the T1069C polymorphism in the SNAP-25 gene and the ADHD in the Portuguese population, it was proceeded to a sample collection of probands and their parents (trios). Genotyping was performed by PCR–RFLP and the resulting data were analyzed according to HRR and TDT strategies. The results showed a trend of association between the T1069C polymorphism of SNAP-25 gene and ADHD (HRR: c2=3, 50; d.f =1; p=0,061; TDT: c2=3, 75; d.f =1, p=0,053). Thus, the association strategy based on trios and HRR and TDT methods, which revealed a trend of association between the T allele of the T1069C polymorphism of SNAP-25 gene and the ADHD, seems to suggest that the identified polymorphism may play a direct role in the etiology of ADHD, in the Portuguese population. It will be important to use a larger sample, as well as the analysis of haplotypes, to clarify a possible association between the SNAP-25 gene and the ADHD, in the Portuguese population |
