Title: Oxiesteróis citotóxicos: síntese e avaliação biológica

Abstract: Os oxiesteróis representam um grupo de lípidos derivados do colesterol que tem atraído muita atenção nos últimos anos devido à sua relevante bioactividade, que varia de acordo com alterações estruturais observadas no núcleo esteróide. Os oxiesteróis estão implicados numa série de importantes processos biológicos, como por exemplo na regulação da homeostase do colesterol, inflamação e diferenciação celular, no entanto, foi o seu perfil citotóxico que mais despertou o nosso interesse, focado em sintetizar, isolar e avaliar a bioactividade destas moléculas. No presente dissertação, foi sintetizada uma biblioteca de novos derivados oxiesteróis acetalizados nos anéis A e B do núcleo esteróide, com o objectivo de obter novas moléculas que induzam a toxicidade celular em células cancerígenas. Nos processos de síntese efectuados, ao longo de todo o procedimento, o custo, a toxicidade e a estabilidade dos reagentes usados foram tidos em conta, procurando seguir os princípios da química verde. As reacções efectuadas foram rápidas, simples e apresentaram bons rendimentos. No presente estudo relatamos ainda a avaliação in vitro de seis novos oxiesteróis em células humanas de cancro do pulmão e estabelecemos estudos de relações estruturaactividade. A actividade anti-proliferativa destes novos compostos foi comprovada quando se verifica uma alteração estrutural no anel B, que envolve uma acetalização e a presença de um hidroxilo na posição 3β do colestano. Os resultados obtidos revelaram a actividade citotóxica dos oxiesteróis a concentrações micromolares de uma forma dose-dependente. Por outro lado, comprovamos também que a acetalização do esteróide no anel A não é uma opção a ter em linha de conta. Esta série de derivados dos oxiesteróis não apresentou actividade citotóxica relevante. Em suma, dependendo das características estruturais dos oxiesteróis, podemos afirmar que alguns destes compostos poderão ser candidatos para desenvolvimento com vista à sua utilização na terapia do cancro.
The oxysterols represent a cholesterol-derived lipid group that has gained significant interest in the last few years due to its own bio-activity relevance, which changes according to structural modifications observed in the steroid core. This group of steroids is implicated in a serie of important biological processes, such as cholesterol homeostasis regulation, inflammation and cellular differentiation. However, it is their cytotoxic characteristics that has justifies the interest to synthesize, isolate and evaluate the bio-activity of these molecules. In the present work, a library of new acetal oxiesterol derivatives in rings A and B of the steroid core was synthesized with the goal of generating new molecules that induce the cell toxicity in cancer cells. During the synthetic processes develops, the reagents were use, thinking in their cost account, toxicity and stability, considering the principles of green chemistry. The reactions developed were quick, simple and with good yields. The present study also comprises the evaluation in vitro of six new oxysterols in lung cancer applied to human cells and the analysis of structure–activity relationships. The antiproliferative activity of these new compounds was proven when a structural modification in the B-ring, involving an acetalization, and the presence of a hydroxyl group in the 3β position of the colestane has occurred. The results obtained revealed a cytotoxic activity at micromolar concentrations and in a dose-dependent form. On the other hand, it was also proven that the acetalization of the steroid on the Aring is not an option to be considered for further anti-cancer therapies. As a concluding remark, depending on the oxysterols structural features, one can say that some of these compounds can be viable candidates to develop for anti-cancer drugs.
