dc.description.abstractOs inibidores de proteinases encontram-se amplamente distribuídos no reino das plantas, particularmente em sementes e outros órgãos de reserva. Apesar de existir uma vasta literatura sobre a ocorrência de inibidores de proteinases serínicas, cisteínicas e metaloproteinases, há uma lacuna significativa na literatura sobre inibidores naturais de proteinases aspárticas. No entanto, as proteinases aspárticas são uma classe de enzimas que se encontra amplamente distribuída na natureza e tem sido implicada em vários processos fisiológicos e patológicos, como a hipertensão, a SIDA, a doença de Alzheimer e a infecção por microrganismos patogénicos. A raridade de inibidores naturais desta classe de proteinases pode ser devido ao facto de a maior parte das proteinases aspárticas ser sintetizada já numa forma inactiva (zimogénio) por si só, que é apenas clivada e activada quando necessário. Assim sendo, o primeiro objectivo deste trabalho consistiu na identificação, purificação e caracterização de inibidores de proteinases aspárticas em extractos de sementes dormentes de Cynara cardunculus L. Para isso foi criada a melhor abordagem possível de extracção, seguida de análise do conteúdo proteico, por técnicas de PAGE, e da actividade proteolítica e inibitória dos extractos, por técnicas de zimografia e ensaios enzimáticos. Após detecção de actividade inibitória da pepsina nos extractos de sementes, procedeu-se à purificação das fracções inibitórias do extracto desnaturado, por cromatografia de exclusão molecular seguida de cromatografia de fase reversa. As fracções recolhidas foram analisadas para actividade inibitória da pepsina, sendo que as fracções da exclusão molecular inibiram significativamente a enzima, enquanto as fracções da fase reversa não apresentaram qualquer inibição. As fracções da exclusão molecular apresentaram também actividade inibitória contra outras proteinases aspárticas, como a quimosina e a cardosina A. Em termos de conteúdo proteico, nenhuma das fracções apresentou bandas proteicas em SDS-PAGE e Tricina-SDS-PAGE, indicando que as moléculas responsáveis pela inibição ou são péptidos pequenos, que não são detectados pelas técnicas de PAGE, ou são moléculas de natureza proteica. XII De modo a caracterizar parte do conteúdo das fracções inibitórias, estas foram sujeitas a uma cromatografia bidimensional de camada fina, tendo-se detectado a presença de fracções lipídicas com características amino- e glicolipídicas. Estas foram isoladas e separadas por cromatografia de fase reversa, tendo-se obtido um perfil semelhante para ambas as fracções, o que indica que podemos estar na presença de moléculas glicolipídicas incorporadas com resíduos aminoácidos. O segundo objectivo do trabalho consistiu na avaliação da capacidade antimicrobiana das fracções inibitórias purificadas do extracto desnaturado. Para isso foram realizados ensaios antimicrobianos, pelo método de difusão em discos, contra estirpes patogénicas de bactérias (Escherichia coli e Staphylococcus aureus) e de leveduras (Candida albicans e Saccharomyces cerevisae). dc.description.abstractProteinase inhibitors are widely distributed among the plant kingdom, particularly in seeds and other reserve organs. Regardless of the extensive literature about the occurrence of serinic, cysteinic and metallo-proteinases inhibitors, there is a significant gap in the literature about naturally occurring aspartic proteinase inhibitors. However, aspartic proteinases are a class of enzymes that is widely distributed in nature and has been implicated in several patho- and physiological processes, such as hypertension, AIDS, Alzheimer’s disease and infections by pathogenic microorganisms. The rarity of naturally occurring inhibitors of this proteinases’ class can be explained by the fact that most of the aspartic proteinases are synthesized by itself in a inactive form (zymogen), that is only cleaved and activated when necesssary. Therefore, the first aim of this work was the identification, purification and characterization of aspartic proteinase inhibitors in Cynara cardunculus L. dormant seed extracts. Thereunto, it was created the best possible extraction approach, followed by analysis of protein profile, by PAGE techniques, and proteolytic and inhibitory activity of the extracts, by zymography techniques and enzymatic assays. After detection of inhibitory activity against pepsin in the seed extracts, the next step was the purification of the inhibitory fractions of the denaturated extract, by molecular exclusion chromatography followed by reverse-phase chromatography. The collected fractions were tested for inhibitory activity against pepsin: molecular exclusion fractions significantly inhibited pepsin, while reverse-phase fractions showed no inhibition of this enzyme. The molecular exclusion fractions also showed significant inhibitory activity toward other aspartic proteinases, namely chymosin and cardosin A. In terms of protein profile, none of the fractions showed protein bands in SDSPAGE or Tricina-SDS-PAGE, meaning that the molecules responsible for the inhibition are either small peptides, which are not detected by the PAGE techniques, or molecules of non-protein nature. In order to characterize part of the inhibitory fractions’ content, these were applied to a bidimensional thin-layer chromatography, where it was detected the presence of lipidic fractions with amino- and glycolipid features. These lipidic fractions were isolated and separated by reverse-phase chromatography, where the profile was XIV similar for both fractions, indicating that we can be in the presence of glycolipidic molecules incorporated with amino acids residues. The second aim of this work was the evaluation of the inhibitory fractions’ antimicrobial ability. Thereunto, antimicrobial assays were carried out by the disk diffusion method, against pathogenic bacterial (Escherichia coli and Staphylococcus aureus) and yeast (Candida albicans and Saccharomyces cerevisae) strains.
