síntese de novos compostos heterocíclicos funcionalizados via reacções de cicloadição.
Este objectivo foi alcançado através do estudo de reacções de ciclo-adição 1,3-
dipolar de alenoatos, na qual estes podem funcionar como percusores de dipolos e
como dipolarófilos. Uma outra vertente estudada foi a reacção de hetero-Diels-Alder
de novos nitroso-alquenos e azo-alquenos contendo um grupo tetrazole na posição 3.
Numa primeira aproximação foram realizados estudos da utilização de
alenoatos para a preparação de nitronas e subsequente ciclo-adição 1,3-dipolar.
Reacções de adição de N-metil-hidroxilamina a buta-2,3-dienoato de benzilo, penta-
2,3-dienoato de benzilo e 4-fenilbuta-2,3-dienoato de benzilo permitiram a síntese
eficiente das correspondentes nitronas. A ciclo-adição 1,3-dipolar em condições suaves
destes dipolos com alquinos e buta-2,3-dienoato de benzilo originou a síntese de
várias 4-isoxazolinas. Por outro lado, a utilização de condições de reacção mais severas
conduziu à formação dos correspondentes pirróis.
Com o objectivo de esclarecer o mecanismo envolvido no rearranjo de 4-
isoxazolinas nos correspondentes pirróis foram efectuados cálculos computacionais.
Estes estudos fundamentaram a proposta mecanística para este rearranjo, envolvendo
a formação de 2-acil-aziridinas como intermediário. O isolamento dum 3-hidroxi-2,3-
di-hidro-1H-pirrole numa das reacções estudadas confirmou igualmente a formação
destes compostos como outro intermediário da isomerização de 4-isoxazolinas em
pirróis.
A procura de novos e eficientes métodos para a obtenção de 3-pirrolinas e
ciclopentenos levou-nos a explorar a ciclo-adição [3+2] formal de alenoatos catalisada
por fosfinas sob irradiação de micro-ondas e com recurso a catalisadores
heterogéneos. As condições convencionais de reacção levaram à síntese das 3-
pirrolinas e ciclopentenos com bons rendimentos à temperatura ambiente, enquanto
que a reacção induzida por micro-ondas levou igualmente à síntese dos ciclo-aductos
com rendimentos semelhantes, mas reduzindo significativamente o tempo de reacção
para apenas 5 minutos. Por outro lado, a utilização de fosfinas em suporte sólido na
reacção de ciclo-adição de buta-2,3-dienoatos, conduziu à formação dos ciclo-aductos
pretendidos, com vantagem de recuperação e reutilização dos catalisadores.
Foi desenvolvida uma estratégia sintética para a obtenção de novos N-vinilheterociclos
de forma regio- e estereosselectiva de heterociclos, derivados de amino
ésteres. A adição de Michael de cis-3-fenil-aziridina-2-carboxilato de etilo ao buta-2,3-
dienoato de benzilo originou a correspondente N-vinil-aziridina. A abertura
conrotatória do anel aziridina induzida por micro-ondas gerou o correspondente ileto
de azometino que foi interceptado em ciclo-adições 1,3-dipolares dando N-vinilpirrolina,
N-vinil-pirrolidinas e N-vinil-triazolina. Uma segunda vertente do estudo descrito nesta dissertação centrou-se no
desenvolvimento de estratégias sintéticas para a obtenção de heterociclos substituídos
com um ácido tetrazólico via reacção de Diels-Alder de 3-(1H-tetrazol-5-il)-nitrosoalquenos
e 3-(1H-tetrazol-5-il)-1,2-diazabuta-1,3-dienos. 1H-Tetrazóis substituídos na
posição 5 são isósteros não clássicos do grupo carboxílico, sendo mais lipofílicos e
exibindo maior resistência aos mecanismos metabólicos de degradação biológica,
características que podem conduzir ao aumento de actividade biológica.
Neste contexto, foram efectuados estudos para a geração de 3-(1H-tetrazol-5-
il)-nitroso-alquenos e reactividade em reacções de hetero-Diels-Alder com alquenos
ricos em electrões e heterociclos aromáticos de cinco membros. Esta metodologia
levou à síntese de uma nova classe de compostos que combinam a estrutura de 1,2-
oxazinas e oximas de cadeia aberta com um substituinte tetrazólico. A existência dum
grupo substituinte facilmente removível em N-1 do tetrazole abriu a possibilidade de
após a reacção de ciclo-adição ser efectuada a desprotecção. Por outro lado, a redução
de oximas de cadeia aberta originou a síntese de uma nova classe de compostos que
após desprotecção do tetrazole permitiu a formação de 5-(1-amino-alquil)-1Htetrazóis,
bioisósteros de α-amino ácidos.
A reacção de Diels-Alder do tipo inverso de 3-(1H-tetrazol-5-il)-1,2-diazabuta-
2,3-dienos foi igualmente explorada. Estes dienos participam na reacção de cicloadição
como componente 4 com dienófilos deficientes e ricos em electrões e com
heterociclos aromáticos de 5 membros, originando 1,4,5,6-tetra-hidropiridazinas e
hidrazonas com bons rendimentos de forma regio- e estereosselectiva. A presença de
um grupo protector no tetrazole permitiu, após reacção de ciclo-adição [4+2] com os
diferentes dienófilos, a sua desprotecção para obtenção de tetra-hidropiridazinas
substituídas na posição 3 com um grupo ácido tetrazólico.
Os resultados alcançados permitiram um conhecimento mais aprofundado da
química de alenoatos, de 3-terazolil-nitroso-alquenos e 3-tetrazolil-1,2-diazabuta-1,3-
dienos. O estudo da reactividade destes valiosos intermediários sintéticos conduziu à
síntese de diversos compostos heterocíclicos funcionalizados com elevado interesse a
nível químico e biológico. The main purpose of the work presented in this dissertation was the synthesis of new functionalized heterocyclic compounds via cycloaddition reactions. This was achieved by studying 1,3-dipolar cycloaddition reactions of allenoates in which they can function as dipole precursors and as dipolarophiles. Another aspect studied was the hetero-Diels-Alder reaction of new nitrosoalkenes and azoalkenes containing a tetrazole group in position 3. In a first approximation studies were performed using allenoates to prepare nitrones and subsequent 1,3-dipolar cycloaddition. Addition reactions of Nmethylhydroxylamine to benzyl buta-2,3-dienoate, benzyl penta-2,3-dienoate and benzyl 4-phenylbuta-2,3-dienoate allows the efficient synthesis of the corresponding nitrones. 1,3-Dipolar cycloaddition of these dipoles with alkynes and benzyl buta-2,3- dienoate under mild conditions originated several 4-isoxazolines. Moreover, the use of more severe reaction conditions led to the formation of the corresponding pyrroles. In order to clarify the mechanism involved in the rearrangement of the 4- isoxazolines into the corresponding pyrroles, quantum chemical calculations have been performed. These studies substantiate the proposed mechanism for this rearrangement, involving the formation of 2-acylaziridine as intermediate. The isolation of a 3-hydroxy-2,3-dihydro-1H-pyrrole in one of the reactions studied also confirmed the formation of these compounds as another intermediate of the 4- isoxazoline pyrrole isomerization. The search for new and efficient methods for obtaining 3-pyrrolines and cyclopentenes led us to explore the phosphine-catalysed formal [3 +2] cycloaddition of allenoates under microwave irradiation and using of heterogeneous catalysis. Conventional reaction conditions led to the synthesis of 3-pyrrolines and cyclopentenes in good yields at room temperature, while the reaction induced by microwaves also led to the synthesis of the same cycloadducts with similar yields, but significantly reducing the reaction time to only 5 minutes. Moreover, the use of phosphines on solid support in the cycloaddition reaction of buta-2,3-dienoates led to the formation of desired cycloadducts, with the advantage of recovery and reuse of the catalyst. We developed a synthetic strategy for the synthesis of new N-vinyl heterocycles, in a heterocyclic regio- and stereoselective manner, from amino esters. The Michael addition of ethyl cis-3-phenylaziridine-2-carboxylate to benzyl buta-2,3- dienoate gave the corresponding N-vinylaziridine. The conrotatory ring opening of the aziridine induced by microwaves generates the corresponding azomethine ylide which was trapped in 1,3-dipolar cycloadditions giving N-vinylpyrroline, N-vinylpyrrolidines, and N-vinyltriazoline. A second aspect of the study described in this work has focused on developing synthetic strategies for obtaining substituted heterocycles with a tetrazolic acid moiety via Diels-Alder reaction of 3-(1H-tetrazol-5-yl)-nitrosoalkenes and 3-(1H-tetrazol-5-yl)- 1,2-diazabuta-1,3-dienes. 1H-Tetrazoles substituted in position 5 are effective bioisosteres of the carboxyl group, being more lipophilic and exhibiting stronger resistance to metabolic degradation mechanisms, characteristics that can lead to increased of biological activity. In this context, studies have been performed on the generation of 3-(1Htetrazol- 5-yl)-nitrosoalkenes and reactivity in hetero-Diels-Alder reactions with electron-rich alkenes and aromatic heterocycles with five members. This approach led to the synthesis of a new class of compounds which combine the structure of 1,2- oxazines and open chain oximes bearing a 1H-tetrazolyl substituent. The existence of an easily removable substituent group in N-1 of the tetrazole, makes deprotection possible after the cycloaddition reactions. Moreover, reduction of oximes open chain originated the synthesis of a new class of compounds which, after deprotection of the tetrazole, allowed the formation of 5-(1-aminoalkyl)-1H-tetrazoles, bioisosteres of α- amino acids. On the other hand, the Diels-Alder reaction with inverse electronic demand of 3-(1H-tetrazol-5-yl)-1,2-diazabuta-2,3-dienes has also been explored. These dienes participate in the cycloaddition reaction as 4 component with electron-deficient and electron-rich alkene dienophiles and aromatic 5-membered heterocyclic dienophiles, yielding 1,4,5,6-tetrahydropyridazines and hydrazones with good yields in a regio- and stereoselective manner. The presence of a protecting group on the tetrazole allowed, after [4+2] cycloaddition with different dienophiles, its deprotection to obtain the tetrahydropyridazines substituted in position 3 with an acid tetrazole moiety. The results obtained allow a better understanding of the chemistry of allenoates, 3-tetrazolyl-nitrosoalkenes and 3-tetrazolyl-1,2-diazabuta-1,3-dienes. The study of the reactivity of these valuable synthetic intermediates led to the synthesis of various functionalized heterocyclic compounds with high chemical and biological interest. |
