dc.description.abstract | Psoriasis, one of the most prevalent inflammatory skin diseases in humans, arises from a complex interplay between innate and adaptive immune cells, resulting in persistent inflammation. This condition is a product of genetic and environmental factors, leading to a spectrum of symptoms ranging from mild discomfort to severe manifestations, significantly affecting the individual's quality of life.A central element in the skin's immunoinflammatory response is represented by keratinocytes, a type of skin cell. The cells are key players in the pathogenesis of psoriasis, actively participating in both the initiation and maintenance phases of the condition.This thesis delves deeply into the molecular aspects of psoriasis, with a specific emphasis on the protein CARD14 and, more precisely, its splice variant CARD14sh. This variant exhibits increased expression in the skin and is closely associated with psoriasis development. Despite its importance, the regulatory mechanisms involving CARD14sh have remained poorly understood. Moreover, various mutated forms of CARD14sh have been found in psoriasis patients, motivating our investigation into the impact of these mutations on psoriasis-related cellular characteristics.Our research delves into the impact of psoriasis-associated CARD14sh mutants on diverse keratinocyte morphological characteristics evident in psoriasis. Furthermore, we explore the modulation of the TBK1/IRF3 signalling pathway by CARD14sh in keratinocytes, unveiling its suppressive effect on P-IRF3 levels and its potential implications for psoriasis pathogenesis. In addition to these significant findings, our study reveals a novel interaction between CARD14sh and MAVS in keratinocytes, offering fresh insights into the underlying regulatory mechanisms that contribute to the development of this disorder.Overall, our data significantly advances the understanding of the role of CARD14sh in the molecular mechanisms underlying inflammatory skin disorders such as psoriasis. | eng |
dc.description.abstract | A Psoríase, uma das doenças inflamatórias de pele mais prevalentes em seres humanos, surge de uma complexa interação entre células imunes inatas e adaptativas, resultando numa inflamação persistente. Esta condição é resultado da combinação de fatores genéticos e ambientais, causando um espectro de sintomas que vão desde desconforto leve até manifestações graves, afetando significativamente a qualidade de vida do indivíduo.Um elemento central na resposta imuno-inflamatória da pele é representado pelos queratinócitos, um tipo de célula da pele. Essas células desempenham um papel fundamental na patogénese da psoríase, participando ativamente tanto na fase de início como na fase de manutenção desta condição.Esta tese mergulha profundamente nos aspetos moleculares da psoríase, com ênfase específico na proteína CARD14 e, mais precisamente, na variante de “splice” CARD14sh. Esta variante mostra uma expressão aumentada na pele e está intimamente associada ao desenvolvimento da psoríase. Apesar de sua importância, os mecanismos regulatórios envolvendo a CARD14sh têm permanecido pouco compreendidos. Além disso, várias formas mutantes de CARD14sh têm sido encontradas em pacientes com psoríase, motivando nossa investigação sobre o impacto dessas mutações em características celulares relacionadas esta patologia.A nossa pesquisa explora o impacto dos mutantes CARD14sh associados à psoríase em diversas características morfológicas dos queratinócitos evidentes na psoríase. Além disso, investigamos a modulação da via de sinalização TBK1/IRF3 pelo CARD14sh nos queratinócitos, revelando o seu efeito supressor nos níveis d P-IRF3 e as suas potenciais implicações na patogénese da psoríase. Para além destes achados significativos, o nosso estudo revela uma interação inédita entre o CARD14sh e o MAVS nos queratinócitos, oferecendo novos conhecimentos sobre os mecanismos regulatórios subjacentes que contribuem para o desenvolvimento desta doença.Em geral, nossos dados avançam significativamente na compreensão do papel da CARD14sh nos mecanismos moleculares subjacentes a distúrbios inflamatórios da pele, como a psoríase. | por |
