dc.description.abstract | A Artrite Reumatoide (AR) é uma doença caracterizada por inflamação crónica, sendo a doença reumática mais comum tendo uma prevalência de 1% na população mundial. É uma doença que afeta predominantemente mulheres (com uma proporção de 3:1). É considerada uma doença multifatorial uma vez que a sua etiologia passa não só por fatores genéticos, mas também por fatores ambientais. Devido aos fatores acima descritos, a investigação mais aprofundada desta doença, torna-se extremamente importante. A patologia da doença está intimamente relacionada com a presença de células T e recentemente o laboratório de acolhimento descreveu um subtipo de células T presentes no líquido sinovial e sangue de doentes com RA, que para além de apresentar características típicas de células T folicular auxiliar possui Recetor do tipo Toll 4 na sua superfície. Foram denominadas células TLR4+ Tfh-“like”. As características acima mencionadas atribuem-lhes a capacidade de produzir IL-21 levando à produção de anticorpos, bem como à produção de IL-17 que promove a inflamação. Colocámos a hipótese de que estas células poderiam ter um perfil citotóxico associado, com implicações, por exemplo, na morte de osteoblastos. Para demonstrar a nossa hipótese, utilizámos células de sangue periférico de pacientes com Artrite Reumatoide, de forma a realizar uma caracterização fenotípica. Desta forma, foi investigada a eventual presença de Ligando Fas (FasL) e “Natural-Killer Group 2, member D” (NKG2D), ambas moléculas citotóxicas. Foi possível concluir, através deste estudo, que se verifica a expressão de FasL e NKG2D à superfície das células TLR4+ Tfh-like o que sugere o seu potencial citotóxico. | por |
dc.description.abstract | Rheumatoid arthritis (RA) is a disease characterized by chronic inflammation and is the most common rheumatic disease with a prevalence of 1% in the world population. It is a disease that predominantly affects women (with a ratio of 3:1). The disease is considered to be multifactorial, since its etiology involves not only genetic but also environmental factors. Due to the abovementioned factors, further investigation of this disease becomes extremely important. The pathology of the disease is closely related to the presence of T cells. Recently the hosting laboratory described a subset of T cells present in the synovial fluid and blood of patients with RA, which in addition to presenting typical characteristics of follicular helper T cells has Toll-like receptor 4 on its surface. They were named TLR4+ Tfh-like cells. The above-mentioned characteristics give them the ability to produce IL-21 leading to antibody production as well as IL-17 production which promotes inflammation. We hypothesized that these cells may have an associated cytotoxic profile with implications for example in osteoblast death. To address our hypothesis, blood donated by Rheumatoid Arthritis patients was used to perform a phenotypic characterization in which the possible presence of Fas ligand (FasL) and Natural-Killer Group 2, member D (NKG2D), both cytotoxic molecules, was investigated. Through this study, it was possible to conclude that the expression of FasL and NKG2D on the surface TLR4+ Tfh-like cells is verified, which, by itself, does not prove their cytotoxic profile, but suggests their potential. | eng |
