dc.description.abstract | As tauopatias compreendem mais de vinte doenças neurodegenerativas, incluindo a doença de Alzheimer. Atualmente, a doença de Alzheimer é a forma mais comum de demência e representa um grande problema socioeconómico para a nossa sociedade, afetando cerca de 47 milhões de pessoas em todo o mundo. A doença de Alzheimer é caracterizada pela deposição de placas de amiloide e acumulação de tau no cérebro, levando a um declínio cognitivo gradual. Ao longo dos anos, tem sido feitos avanços significativos na investigação da doença de Alzheimer, no entanto, ainda não existe um tratamento eficaz para esta doença. As intervenções farmacológicas disponíveis consistem no uso de medicamentos que podem aliviar alguns sintomas em doentes com doença de Alzheimer. Grandes avanços na compreensão da genética e patogénese molecular da doença de Alzheimer permitiram a entrada de várias moléculas direcionadas contra a beta-amiloide e tau para os ensaios clínicos. No entanto, imunoterapias ativas e passivas contra a beta-amiloide falharam em demonstrar evidências que suportem a eliminação da formação de oligomeros pré-existentes. Em alternativa, foi colocada a hipótese de que a identificação de novos alvos que permitam modular a tau patológica poderá constituir uma estratégia viável para combater muitas tauopatias. Recentemente, foi descoberto um alvo promissor contra a tau patológica. A redução dos níveis de expressão de Synaptogyrin-3 (syngr-3) demonstrou resgatar os défices cognitivos resultantes de alterações na tau. dc.description.abstract | Tauopathies comprise over twenty neurodegenerative diseases, including Alzheimer’s disease. Currently, Alzheimer's disease is the most common form of dementia and represents a major socioeconomic burden to our society affecting close to 47 million people worldwide. Alzheimer's disease is characterized by the deposition of amyloid plaques and abnormal tau accumulation in the brain, leading to gradual cognitive decline. Over the years, significant strides were made in Alzheimer's disease research, however there is still no effective treatment for Alzheimer's disease. The available pharmacological interventions rely on the use of drugs that can relieve some of the symptoms in Alzheimer's disease patients. Major breakthroughs in understanding Alzheimer's disease genetics and molecular pathogenesis prompted the intensification of several molecules targeting amyloid-beta and tau to jump into clinical trials. However, active and passive amyloid-beta immunotherapies have failed to show evidence supporting clearance of already existing oligomer formation. Alternatively, identifying novel targets that can modulate pathological tau is predicted to be a feasible strategy to tackle many tauopathies. Recently, a prominent target for pathological tau was discovered. Lowering Synaptogyrin-3 (syngr-3) expression levels were found to rescue cognitive deficits in the tau background.
