Estudo comparativo de diferentes suportes de colheita para amostras biológicas.

Abstract

The collection of forensic evidence and the extraction of the genetic material contained therein are the most relevant steps in the process of determining the genetic profile of an individual.This work aimed to verify the extraction effectiveness of the Casework Direct Kit (PROMEGA), compared to the PrepFilerTM Forensic DNA Extraction (AB) Kit, to verify the compatibility with the chemistry of the reagents involved in the various steps of obtaining a genetic profile, such as differential extraction, orientation and confirmation tests for semen, quantification, amplification, separation and detection of amplified products and optimize protocols for adapting this kit to the routine of forensic laboratories. It also aimed to verify the performance of the nylon swab as a collection device for forensic evidence of a sexual nature, in comparison to the viscose swab used for collection by the INMLCF.The Casework Direct kit proved to be compatible with the orientation (SERATEC PSA Semiquant) and confirmation (Christmas Tree Coloring Technique) tests for semen with the PrepFilerTM Forensic DNA Extraction (the Automate Express Forensic DNA Extraction), Quantifiler Trio DNA Quantification and Powerplex Fusion 6C System. However, it proved to be incompatible with the I-sep differential extraction kit. Although using a simplified and less durable protocol, the Casework Direct Kit is not suitable for forensic samples with a reduced amount of genetic material. The PrepFiler Forensic DNA Extraction kit proved to be more suitable for this type of samples, presenting a much higher yield. The introduction of distilled water optimized the extraction process and the run on the SERATEC cassette. In addition, the mixture of Casework Direct Reagent, distilled water and semen showed viable preliminary tests up to two weeks of storage.Regarding the swabs tested, the nylon swab showed greater efficiency in eluting the genetic material into the solution, compared to the viscose swab.

A colheita de evidências forenses e a extração do material genético nestas contido são os passos mais relevantes do processo de determinação do perfil genético de um indivíduo.Com este trabalho pretendeu-se verificar a rentabilidade de extração do Kit Casework Direct (PROMEGA), comparativamente ao Kit PrepFilerTM Forensic DNA Extraction (AB), verificar a compatibilidade com a química dos reagentes envolvidos nos diversos passos da obtenção de um perfil genético, como a extração diferencial, testes de orientação e confirmação para sémen, quantificação, amplificação, separação e deteção de produtos amplificados e otimizar protocolos para adaptação deste kit à rotina de laboratórios forenses. Teve ainda por objetivo verificar o desempenho da zaragatoa de nylon, como dispositivo de colheita para evidências forenses de natureza sexual, em comparação à zaragatoa viscose utilizada para recolha pelo INMLCF.O Kit Casework Direct demostrou ser compatível com o teste de orientação (SERATEC PSA Semiquant) e confirmação (Técnica de Coloração Christmas Tree) para sémen e com os sistemas PrepFilerTM Forensic DNA Extraction, Quantifiler Trio DNA Quantification e Powerplex Fusion 6C System. Todavia, demostrou ser incompatível com o kit de extração diferencial I-sep. Embora utilize um protocolo mais simplificado e menos duradouro, o Kit Casework Direct não é adequado para amostras forenses com reduzida quantidade de material genético. O Kit PrepFiler Forensic DNA Extraction mostrou ser mais adequado para este tipo de amostras, apresentando um rendimento muito superior. A introdução de água destilada otimizou o processo de extração, e o percurso da amostra na cassete de SERATEC. Além disso, a mistura de Casework Direct Reagent, água destilada e sémen apresentou testes preliminares viáveis até duas semanas de armazenamento.Relativamente às zaragatoas testadas, a zaragatoa de nylon apresentou uma maior eficácia na eluição do material genético para a solução, em comparação à zaragatoa de viscose.
