Abstract: O envelhecimento pode ser definido como um declínio progressivo, com uma perda, dependente da idade, de funções nos sistemas vivos. A senescência é uma resposta celular ao stress e danos graves, caracterizada pelo bloqueio do ciclo celular, regulação positiva das vias pró-sobrevivência e indução de um fenótipo secretor complexo. A senescência celular é um processo ligado à idade, argumentando-se que a percentagem de células senescentes aumenta com esta, aumentando a carga de células senescentes. A limpeza de células senescentes mostra-se eficiente na redução dos efeitos senescentes em diversos modelos in vivo. Estudos recentes identificaram vulnerabilidades específicas nas células senescentes, que podem ser exploradas por um novo grupo de medicamentos, os senolíticos. No entanto, apesar das validações de conceito bem-sucedidas usando senolíticos, a sua potencial aplicação translacional é dificultada pelas toxicidades associadas, sendo necessário o desenvolvimento de meios de entrega eficientes.Assim, criámos uma biblioteca de 30 nanopartículas capazes de encapsular Navitoclax, que são dissociadas na presença de SA-β-Gal. A biblioteca foi criada com base nas interações hospedeiro-hóspede entre polímeros à base de ciclodextrina e adamanteno conjugado com galactan. Os níveis de atividade da SA-β-Gal lisossomal são uma das principais características das células senescentes. Assim, utilizamos esta diferença para dissociar a nanopartícula, pois o galactan é um oligossacarídeo degradado na presença de SA-β-Gal, libertando o senolítico apenas em células senescentes. A atividade senolítica da biblioteca de nanopartículas foi obtida por triagem de alto rendimento em células endoteliais senescentes e saudáveis.Em geral, os resultados mostraram que a maioria das nanopartículas da biblioteca melhoraram o efeito senolítico da Navitoclax em comparação com Navitoclax solúvel, trazendo novas possibilidade terapêuticas para a entrega de compostos senolíticos em células senescentes. Estas senoterapias direcionadas representam uma ferramenta promissora para o tratamento e prevenção de inúmeras patologias e doenças relacionadas à idade.
Aging can be defined as a progressive decline, with age-dependent loss of functions on living systems. Senescence is a cellular response to severe stress and damage, characterized by cell cycle arrest, upregulation of pro-survival pathways and induction of a complex secretory phenotype. Cellular senescence is an age-related process, being argued that the percentage of senescent cells increases with age. Clearance of senescent cells has proven efficiency in reducing senescent effects in several in vivo models, and the use of senolytics as a therapy seems a promising tool to fight aging and age-related diseases. Recent research has identified targetable vulnerabilities of senescent cells that can be exploited by a novel group of drugs, senolytics. Despite successful proofs-of-concept using senolytics, their potential translational application is hampered by their associated toxicities, requiring the development of efficient delivery vehicles.Here, we created a small library of 30 nanoparticles encapsulating Navitoclax, that are dissociated in the presence of senescence-associated β-galactosidase (SA-β-Gal). The library was created based in the host-guest interactions between cyclodextrin-based polymers and adamantane conjugated to galactan. The levels of lysosomal β-galactosidase activity is one of the main characteristics of senescent cells when compared with healthy cells. Here, we take advantage of this difference to disassemble the nanoparticle, since galactan is an oligosaccharide degraded in the presence of SA-β-Gal, releasing the senolytic drug only in senescent cells. The senolytic activity of the nanoparticle library was obtained by high-throughput screening against healthy and senescent endothelial cells. Overall, the results from the library screening showed that the majority of the nanoparticles in the library improved senolytic effect of navitoclax when compared with soluble Navitoclax, opening a new therapeutic tool for targeted delivery of senolytic compounds to senescent cells. These targeted senotherapies represent a promising platform for the treatment and prevention of numerous pathologies and age-related diseases.
