dc.description.abstractAs alergias são um dos maiores problemas de saúde do século XXI, e têm vindo a aumentar cada vez mais ao longo dos últimos anos. A inalação dos esporos e fragmentos de hifas de Alternaria infectoria e Aspergillus fumigatus pode levar ao desenvolvimento não só de alergias, mas também de infeções fúngicas em pacientes imunocomprometidos. Estas duas espécies de fungos filamentosos, são considerados os maiores patogénios aéreos responsáveis por diversas doenças fúngicas.O desenvolvimento de respostas alérgicas no hospedeiro é devido à exposição deste a alergénios da parede celular dos esporos e a alergénios citoplasmáticos, como proteínas intracelulares. Já foram identificadas algumas destas proteínas intracelulares que têm um papel na resposta alérgica. Sendo a parede celular dos fungos uma estrutura rígida e robusta, levantamos a hipótese de que as vesículas extracelulares (EVs) libertadas por estes fungos funcionem como veículo de transporte de alergénios intracelulares para fora da célula fúngica. Efetivamente, alguns estudos demonstram que as EVs de fungos são ativadoras in vitro do sistema imunitário, uma vez que transportam fatores de virulência, antigénios e alergénios.O principal objetivo deste estudo é perceber o papel imunomodelador das EVs quando estas entram em contato com células A549, uma linha celular de epitélio do trato respiratório. Para tal, previamente, procedeu-se a recolha, isolamento e caraterização das EVs. Quantificou-se a proteína total das EVs, percebendo-se que após uma semana, esta degradava, mesmo quando mantidas a 4 ºC. A concentração e tamanho das EVs foram determinados por nanoparticle tracking analysis (NTA), obtendo-se um tamanho de 103,6 ± 1,7 nm para as EVs de A. infectoria e de 105 ± 2,8 nm para as EVs de A. fumigatus e uma concentração na ordem dos 107-108 partículas/mL. A observação da EVs por microscopia eletrónica de transmissão (TEM) permitiu observar a bicamada lipídica das EVs; por está técnica os tamanhos destas são de 97,7 ± 3,4 nm para as EVs de A. infectoria e 94,2 ± 4,6 nm para as EVs de A. fumigatus. As EVs foram marcadas com anticorpo anti-melanina, para identificar nestas a presença de melanina.Antes dos ensaios de interação, a citotoxicidade das EVs nas células A549 foi avaliada aos 15 min, 1 h e 6 h, pelo método do Trypan Blue. Os resultados mostram que, a estes tempos, as EVs não apresentam citotoxidade para estas células. Por último, procedeu-se aos ensaios de interação. Após 15 min e 1 h de interação, quantificou-se a expressão de 4 genes, TNF-alpha, TSLP, IFN-gamma e IL-18. Os resultados não mostraram um aumento elevado e abrupto da expressão destes genes, sendo que para o TSLP e para o IFN-gamma não se obteve qualquer expressão. Pode-se concluir que as EVs produzidas por estes dois fungos não induzem uma resposta inflamatória aguda em células epiteliais respiratórias, mas antes uma resposta progressiva e crónica.por
dc.description.abstractAllergies are one of the major health problems of the 21st century and have been increasing over the last years. Spores and hyphal fragments inhalation of Alternaria infectoria and Aspergillus fumigatus can trigger not only allergies but may also lead to fungal infections in immunocompromised patients. These two species of filamentous fungi are considered the major airborne pathogens responsible for causing fungal diseases.The orchestration of allergic responses in the host is caused by cell wall spore allergens but also by fungal cytoplasmatic allergens, as intracellular proteins. Some of these intracellular proteins that have a role in allergic response have been identified. Considering the cell wall is a rigid and robust structure, we aim to appraise if the EVs released by these fungi functions as a transport vehicle of allergic components out of the fungal cell. Some studies support that fungal EVs are in vitro activators of immune system, since they carry virulence factors, antigens and allergens.The main objective of this study is to understand the immunomodulatory effect of fungal EVs when they come into contact with the A549 human respiratory epithelial cells. To achieve this, fungal EVs were isolated and characterized. The protein content was quantified, and we realized that the protein was totally lost after one week at 4 ºC. The size and concentration of the obtained EVs was measured by nanoparticle tracking analysis (NTA), with 103.6 ± 1.7 nm for A. infectoria EVs and 105 ± 2.8 nm for A. fumigatus EVs and a concentration of 107-108 particles/mL. By transmission electron microscopy (TEM), it was possible to observe the EVs lipidic bilayer and the size measured by this technique was 97.7 ± 3.4 nm for A. infectoria EVs and 94.2 ± 4.6 nm for A. fumigatus EVs. To identify the presence of melanin in the EVs, these were labelled with an antibody anti-melanin.Before the interaction assays, the cytotoxicity of the EVs on A549 cells were evaluated at 15 minutes, 1 h and 6 h, using the Trypan Blue test. The results obtained show that EVs are not cytotoxic to these cells. Then, we exposed A549 cells to fungal EVs freshly prepared and characterized to appraise if these EVs cause alterations in the TNF-alpha, TSLP, IFN-gamma and IL-18 gene expression after 15 minutes and 1 h of interaction. This interaction caused a discrete modification of the expression of TNF-alpha and IL-18 genes; no expression of the genes coding for TSLP and IFN-gamma was registered.It could be concluded that A. fumigatus and A. infectoria EVs do not trigger an acute pro-inflammatory response by A549 respiratory epithelial cells, but rather a progressive and chronic response.eng
