dc.description.abstractA leucemia linfoblástica aguda (LLA) é uma neoplasia hematológica que afeta a linhagem de linfócitos T e B caracterizada por excessiva proliferação de células linfoide imaturas que invadem a medula óssea, o sangue periférico e outros órgãos linfoides. As alterações genéticas, que incluem aneuploidias, rearranjos cromossómicos, deleções e mutações na sequência do DNA, contribuem para o desenvolvimento e progressão da LLA. Além destas alterações genéticas/cromossómicas, o stresse oxidativo também foi detetado em várias neoplasias hematológicas, incluindo leucemias agudas e crónicas. Na LLA, há um aumento da produção de espécies reativas de oxigénio (ROS) e uma diminuição das defesas antioxidantes, o que evidência o envolvimento do stresse oxidativo na doença.A via KEAP1-NRF2 é um importante mediador da resposta citoprotetora ao stresse exógeno e endógeno induzido pelas ROS. O NRF2 (Nuclear factor erythroid-2-related factor 2) é um fator de transcrição que desempenha um papel crucial na defesa celular contra o stresse oxidativo através da ativação de diversos genes citoprotetores e destoxificantes. Sob condições fisiológicas, o NRF2 está localizado no citoplasma sendo inibido pela KEAP1 (Kelch-like ECH-associated protein 1). No entanto sob condições de stresse, o NRF2 é libertado do complexo KEAP1-NRF2 e transloca-se para o núcleo. No núcleo, o NRF2 forma um heterodímero com as proteínas MAF e liga-se a sequências ARE (elemento de resposta antioxidante), estimulando a expressão dos genes alvos.No entanto, o NRF2 desempenha um papel duplo na carcinogénese. Este fator de transcrição protege as células normais da transformação maligna através da ativação dos seus genes alvos, atuando, portanto, como um supressor tumoral. No entanto, a ativação do NRF2 nas células cancerígenas facilita a progressão do cancro e confere resistência à quimio- e à radioterapia.Deste modo, este projeto investigou o potencial terapêutico de moduladores do NRF2 – um inibidor do NRF2 (Brusatol) e um ativador (Oltipraz) – em linhas celulares da LLA-T (células MOLT-4 e CEM) e LLA-B (células KOPN8). No contexto deste projeto, os níveis de expressão génica do NFE2L2, o gene que codifica o NRF2, foram avaliados através da qPCR. As linhas celulares de LLA foram incubadas na ausência e presença de Oltipraz e Brusatol em monoterapia durante 72h. A atividade metabólica foi determinada pelo ensaio de resazurina. O tipo de morte celular foi avaliado por citometria de fluxo recorrendo à dupla marcação com anexina V/ 7-amino-actinomicina D (7-AAD) e por análise morfológica através da coloração de Giemsa. O ciclo celular foi avaliado por citometria de fluxo através da utilização da solução iodeto de propídio(PI)/RNAse. Os níveis de ROS, de glutationa reduzida (GSH) e o potencial de membrana mitocondrial foram quantificados por citometria de fluxo através de sondas fluorescentes DCFH2-DA, DHE, MO e JC-1, respetivamente. Os resultados foram analisados estatisticamente considerando um nível de significância de 95% (p <0,05).Os resultados demonstraram que as células MOLT-4 apresentam níveis de expressão mais elevados do gene NFE2L2 (0,137 ± 0,036) do que as células CEM (0,059 ± 0,004). No entanto, as células KOPN8 (0,121 ± 0,022) apresentam níveis de expressão mais elevados do que as células CEM mas mais baixo que as células MOLT-4. O Oltipraz e o Brusatol reduzem a atividade metabólica de modo dependente da concentração, tempo e linha celular, sendo as células de LLA-B mais sensíveis a ambos os fármacos (KOPN8: Oltipraz IC50 = 4,5 μM, Brusatol IC50 = 1,4 nM) relativamente às células de LLA-T (MOLT-4: Oltipraz IC50 = 87,4 μM; Brusatol IC50 = 7,8 nM; CEM: Oltipraz IC50 = 40,4 μM; Brusatol IC50 = 7,4 nM). Além disso, o Oltipraz não mostrou efeito citotóxico e citostático nas células de LLA-T. No entanto, nas células de LLA-B o Oltipraz 250 μM induziu efeito citostático, com bloqueio do ciclo celular em fase G0/G1. Quando as células de LLA-T foram tratadas com Brusatol 25 nM observou-se efeito citotóxico, principalmente morte celular por apoptose e citostático com bloqueio em fase G0/G1. Em relação às células de LLA-B apenas, se verificou bloqueio do ciclo celular em fase G0/G1.Nas células de LLA-T o Oltipraz induziu aumento dos níveis intracelulares de ROS (peróxidos e anião superóxido) e de GSH, embora não sendo estatisticamente significativo. No entanto, nas células LLA-B ocorreu um aumento dos níveis de ROS e uma diminuição dos níveis de GSH. Quando as células foram tratadas com o Brusatol observou-se aumento dos níveis intracelulares de ROS com uma diminuição nos níveis intracelulares de GSH, exceto nas células de LLA-B onde ocorreu uma diminuição dos níveis de ROS e GSH. Por fim, ambos os fármacos diminuíram o potencial de membrana mitocondrial em todas as linhas celulares estudadas.Em conclusão, os resultados obtidos sugerem que tanto o Oltipraz como o Brusatol podem representar possíveis abordagens terapêuticas para as neoplasias linfoides.por
dc.description.abstractAcute lymphoblastic leukemia (ALL) is a hematological neoplasia that affects the lineage of T and B lymphocytes characterized by excessive proliferation of immature lymphoid cells that invade the bone marrow, peripheral blood and other lymphoid organs. Genetic alterations, which include aneuploidies, chromosomal rearrangements, deletions and mutations, contribute to the development and progression of ALL. In addition to these genetic/chromosomal alterations, oxidative stress has also been detected in several hematological neoplasms, including acute and chronic leukemias. In ALL, there is an increase in the production of reactive oxygen species (ROS) and a decrease in antioxidant defenses, what evidence the involvement of oxidative stress in this disease.The KEAP1-NRF2 pathway is an important mediator of the cytoprotective response to exogenous and endogenous stress induced by ROS. NRF2 (Nuclear factor erythroid-2-related factor 2) is a transcription factor that plays a crucial role in cellular defense against oxidative stress through the activation of several cytoprotective and detoxifying genes. Under physiological conditions, NRF2 is located in the cytoplasm and is inhibited by KEAP1 (Kelch-like ECH-associated protein 1). However, under stress conditions, NRF2 is released from the KEAP1-NRF2 complex and translocates to the nucleus. In the nucleus, NRF2 forms a heterodimer with MAF proteins and binds to ARE sequences (antioxidant response element), stimulating the expression of target genes.However, NRF2 plays a dual role in carcinogenesis. This transcription factor protects normal cells from malignant transformation by activating their target genes, thus acting as a tumor suppressor. However, the activation of NRF2 in cancer cells facilitates the progression of cancer and confers resistance to chemo- and radiotherapy.Thus, this project investigated the therapeutic potential of NRF2 modulators - an NRF2 inhibitor (Brusatol) and an activator (Oltipraz) - in T-ALL (MOLT-4 and CEM cells) and B-ALL cell lines (KOPN8 cells).In the context of this project, the levels of gene expression of NFE2L2, the gene encoding NRF2, were assessed using qPCR. ALL cell lines were incubated in the absence and presence of Oltipraz and Brusatol as monotherapy for 72 hours. Metabolic activity was determined by the resazurin assay. The type of cell death was assessed by flow cytometry using double staining with annexin V/ 7-aminoactinomycin D (7-AAD) and by morphological analysis using Giemsa stain. The cell cycle was evaluated by flow cytometry using the propidium iodide (PI)/RNAse solution. The levels of ROS, reduced glutathione (GSH) and the potential of mitochondrial membrane were quantified by flow cytometry using fluorescent probes DCFH2-DA, DHE, MO and JC-1, respectively.The results were analyzed statistically considering a 95% significance level (p <0.05).The results showed that MOLT-4 cells have higher levels of expression of the NFE2L2 gene (0.137 ± 0.036) than CEM cells (0.059 ± 0.004). However, KOPN8 cells (0.121 ± 0.022) have higher levels of expression than CEM cells but lower than MOLT-4 cells. Oltipraz and Brusatol reduce metabolic activity depending in concentration, time and cell line, with B-ALL cells being more sensitive to both drugs (KOPN8: Oltipraz IC50 = 4.5 µM, Brusatol IC50 = 1.4 nM) for T-ALL cells (MOLT-4: Oltipraz IC50 = 87.4 µM; Brusatol IC50 = 7.8 nM; CEM: Oltipraz IC50 = 40.4 µM; Brusatol IC50 = 7.4 nM). In addition, Oltipraz showed no cytotoxic and cytostatic effect on T-ALL cells. However, in B-ALL cells, 250 µM Oltipraz induced a cytostatic effect, blocking the cell cycle in phase G0/G1. When T-ALL cells were treated with 25 nM Brusatol, a cytotoxic effect was observed, mainly cell death due to apoptosis and cytostatic with G0/G1 phase block. Regarding B-ALL cells only, there was blockade of the cell cycle in phase G0/G1.In T-ALL cells, Oltipraz induced an increase in intracellular levels of ROS (peroxides and superoxide anion) and GSH, although it was not statistically significant. However, in B-ALL cells there was an increase in ROS levels and a decrease in GSH levels. When cells were treated with Brusatol, an increase in intracellular levels of ROS was observed with a decrease in intracellular levels of GSH, except in B-ALL cells where there was a decrease in ROS and GSH levels. Finally, both drugs decreased the mitochondrial membrane potential in all studied cell lines.In conclusion, the results obtained suggest that both Oltipraz and Brusatol may represent possible therapeutic approaches for lymphoid neoplasms.eng
