Abstract: Hoje em dia, o ritmo agitado do dia a dia, é considerado responsável pelo aumento de doenças neurodegenerativas e outras. Como consequência existe um interesse crescente no conhecimento dos mecanismos celulares e o caminho para a descoberta de novas formas de tratamento tornou-se um foco principal de investigação. Por isso, é fundamentalo efeito que muitos compostos induzem no sistema nervoso central.Este trabalho permitiu a medição de variações óticas de autofluorescência e de ROS, usandoluz incidente de 480 nm. Os sinais de ROS verdadeiros foram obtidos subtraindo, ponto a ponto, dados de fatias não incubadas de dados de fatias incubadas com H2DCFDA. A autofluorescência de FAD emitida por estruturas biológicas intrínsecas, quando elas são excitadas por radiação UV/Vis. Ela desempenha um papel importante para a avaliação do estado metabólico celular e da actividade das flavoproteínas mitocondriais, onde o estado oxidativo dos tecidos pode ser verificado. Por outro lado, testar a actividade oxidative em células vivas pode ser bastante desafiante, devido à presença de muitas espécies reativas de oxigénio numa célula apenas. As variações de ROS induzidas foram medidas usando o indicador 2’,7’-diclorodidrofluoresceina diacetato, H2DCFDA. Este corante fluorogénico é usado normalmente para detector o estadooxidativo dentro da célula , formando um composto não fluorescente que é oxidado por ROS, sendo normalmente detetado por espectroscopia de fluorescência. Este trabalho consistiu de experiências focadas essencialmente na caracterização da autofluorescência e da fluorescência associada com com a produção de espécies reactivas de oxigénio (ROS). Os sinais eram induzidos por hipóxia e por estímulos químicos externos, nas sinapses das fibras musgosas da área CA3 do hipocampo de fatias cerebrais de ratos WistarOs agentes estimulantes incluíam o compost despolarizante cloreto de potássio (KCl), o inibidos da enzima mitocondrial citocromo-oxidase azida de sódio (NaN3), o bloqueador de canais de K+ tetraetilamónio (TEA) e o inibidor do canal KATP tolbutamide. O primeiro conjunto de experiências envolveu a aplicação de hipóxia por períodos de 30min e de 60 min, de forma a estudar o efeito da deprivação de oxigénio no metabolism mitochondrial. Nestes estudos, as fatias foram perfundidas com um meio não oxigenado, em equilíbrio com a atmosfera, (21% O2). Usando fatias não incubadas e luz de excitação de 480 nm, observou-se que sinais de autofluorescência, associados com a coenzima mitocondrial FAD, estavam aumentados, provavelmente de uma forma irreversível.. Contudo, os sinais de fluorescência associados com a produção de ROS, revelaram, após serem corrigidos pela componente de autofluorescência, nenhumas ou pequenas variações. A seguir, avaliou-se o efeito de KCl na autofluorescência de FAD e na formação de ROS foi avaliada. Aplicou-se KCl (20 mM) durante períodos de 30 min e de 90 min, para induzir atividade sinática. Além disso,nalgumas experiências, depois de meia hora em KCl, este composto foi aplicado conjuntamente com NaN3 (0.3 mM) para avaliar a influência deste químico tóxico nos mesmos sinais óticos. Os resultados indicam que, e ambas as aplicações de KCl, os sinais de autofluorescência aumentaram, recuperando em grande parte ou totalmente, após a remoção de KCl. Por outrolado, os verdadeiros sinais de ROS exibiram, também em ambos os casos, um pequeno aumento na presença de KCl, seguido de outro aumento para um estado estacionário, após a sua remoção. Por isso, estes sinais não recuperaram. Observou-se um resultado interessante aplicando conjuntamente KCl com NaN3, porque na presença de deste composto, o traço de autofluorescência subiu muito. Contrariamente ao que aconteceu só com KCl, o sinal manteve-se elevado após a saída deste composto. O sinal real de ROS praticamente não tinha variaçõesna presença de NaN3. Contudo, depois da sua retirada, o sinal diminuiupara um nívelinferior à linha base. Finalmente, a ação de outra substância despolarizante, TEA (25 mM), foi também estudada na ausência e presença de tolbutamida (250 M). O objetivo era avaliar o envolvimento de canais KATP nos acontecimentos originados por TEA. Este composto era aplicadoessencialmente durante períodos de 30 min, separadospela perfusão, durante 30 min do fluído cérebroespinal (ACSF). Observou-se que TEA causou uma depressão reversível da autofluorescência e dos sinais de ROS. Na presença de TEA mais tolbutamida, a depressão tinha uma amplitude menor que a devida a TEA apenas. Os resultados estão de acordo com a ideia que a tolbutamida inibe os canias de K,ATP, causando uma despolarização e consequentemente um aumento maior do glutamato e zinco co-libertados. A ação destes neurotransmissor e ião, causa, por sua vez, um aumento de ROS na zona pós-sinática.
Nowadays, the hustle and bustle of everyday life, is found to be responsible for the increasing incidence of neurodegenerative and other diseases. As a consequence, an arising interest in understanding the cellular mechanisms and the path to discover new ways of treatment has become a major research focus. Thus, it is crucial to study the effect that many chemical compounds induce in the central nervous system. This work allowed the measurement of autofluorescence and ROS changes in the optical signals, which were triggered by 480 nm incident light. True ROS signals were obtained as the point-by-point difference, between data from H2DCFDA incubated and non incubated slices. FAD fluorescence is emitted by intrinsic biological structures when they are excited by UV/Vis radiation and play an important role for the evaluation of the metabolic cellular state and of mitochondrial activity of the flavoproteins where the oxidative state of the tissues can be verified. On the other hand, testing oxidative activity in living cells can be really challenging due to the presence of many reactive oxygen species in just one cell. The evoked ROS changes were measured using the 2’,7’-dichlorodihydrofluorescein diacetate indicator, H2DCFDA. This fluorogenic dye is normally used to detect oxidative stress within a cell, forming a non-fluorescent compound which is oxidized by ROS, being usually detected by fluorescence spectroscopy. This work consisted of experiments that were mainly focused on characterizing autofluorescence and fluorescence associated with the production of reactive oxygen species (ROS). The signals were evoked by hypoxia and by external chemical stimuli, at the hippocampal mossy fiber synapses from CA3 hippocampal area of Wistar rat brain slices. The stimulating agents included the depolarizing compound potassium chloride (KCl), the inhibitor of the mitochondrial enzyme cytochrome-oxidase sodium azide (NaN3), the K+ channel blocker tetraethylammonium (TEA) and the KATP channel inhibitor tolbutamide.The first set of experiments involved of the application of 30 min and 60 min periods of hypoxia, in order to study the effect of oxygen deprivation in the mitochondrial metabolism. In these studies, the slices were perfused with a non-oxygenated medium, in equilibrium with atmosphere (21% O2). Using non incubated slices and 480 nm excitation light, it was observed that autofluorescence signals, associated with the FAD mitochondrial coenzyme, were enhanced, most likely in an irreversible way. However, the fluorescence signals associated with ROS production, after correction for the autofluorescence component, revealed none or small changes.Next, the effect of KCl in FAD autofluorescence and in ROS formation was evaluated. KCl (20 mM) was applied during 30 min and 90 min periods, to trigger synaptic activity. In addition, in some experiments, after half an hour in KCl, this compound was combined with NaN3 (0.3 mM) to assess the influence of this toxic chemical in the same optical signals. The results indicate that, in both KCl applications, the autofluorescence signals increased, recovering largely or fully upon KCl washout. On the other hand, the true ROS traces exhibited, also in both cases, a small increase in the presence of KCl, followed by another increase to a steady level, upon its removal. Thus, these signals did not recover.An interesting result was observed when combining KCl with NaN3, since in the presence of this compound, the autofluorescence trace rised very much. Unlike what happened in KCl alone, the signal remained elevated upon washout. The real ROS signal had basically no changes in the presence of NaN3. However, following washout, the signal decreased to a level below the baseline. Finally, the action of another depolarizing drug, TEA (25 mM), was also studied in the absence and presence of tolbutamide (250 M). The goal was to assess the involvement of KATP channels in the TEA evoked events. This compound was mainly applied for periods of 30 min, separated by a 30 min perfusion of the artificial cerebrospinal fluid (ACSF). It was observed that TEA caused a reversible depression of the autofluorescence and ROS signals. In the presence of TEA plus tolbutamide, the depression had a smaller amplitude than the one with TEA alone.The results support the idea that tolbutamide inhibits the KATP channels, causing a depolarization and consequently a higher amount of co-released glutamate and zinc. The action of these neurotransmitter and ion, cause, in turn, an increase in ROS at the postsynaptic site.
