Resumo: A utilização de glifosato tem vindo a aumentar ao longo dos anos, tornando-se num dos herbicidas mais consumidos do mundo. Os seus efeitos no organismo humano e no meio ambiente têm sido analisados e mostram-se preocupantes, à medida que a exposição ao herbicida aumenta. O objetivo do presente estudo foi determinar glifosato em urina de crianças de diversas regiões de Portugal, incluindo zonas urbanas e rurais, para identificar níveis de exposição e a relacionar com os padrões alimentares.Das 41 crianças participantes, 46,34 % (n = 19) eram do sexo masculino, com idades entre 2 e 12 anos, e 53,66 % (n = 22) eram do sexo feminino, com idades entre 2 e 13 anos. A recolha das amostras (urina da primeira micção do dia) ocorreu entre julho de 2018 e fevereiro de 2019. Para além do termo de consentimento, foi preenchido um questionáriopelos pais das crianças, com características antropométricas, hábitos alimentares, estilo de vida e uso de pesticidas. O glifosato foi analisado, através do método ELISA (LOD= 0,6 µg/L) e foi detetado em 95,12 % das amostras, com valores médios de 1,85 ± 0,82 µg/L e valores mínimos e máximos de 0,87 e 4,35 µg/L, respetivamente.Dos 13 estudos mais recentes que determinaram glifosato em urina, apenas 4 utilizaram amostras de crianças, 3 dos quais apresentaram valores superiores aos do presente estudo.No entanto, apenas 1 dos casos apresentou incidência superior à mencionada neste estudo. As diferenças entre as concentrações de glifosato na urina de crianças com idades iguaisou inferiores a 3 anos (1,20 ± 0,32 µg/L), comparativamente às concentrações encontradas em crianças entre os 7 e os 9 anos de idade (2,12 ± 1,00 µg/L) estiveram próximas dasignificância estatistica (p = 0,0631), o que pode ser justificado pelo aumento da atividade da enzima PON1, a partir dos 7 anos de idade. Verificaram-se teores de contaminação superiores em crianças que residiam próximo de zonas agrícolas (<1 km, 2,0 ± 0,97 µg/L), com percentagem superior de consumo de alimentos caseiros (75-100%, 2,72 ± 0,45 µg/L) e cujos pais aplicavam herbicidas no quintal (2,5 ± 1,17 µg/L), quando comparado com o jardim (1,4 ± 0,43 µg/L). Além disso, verificaram-se diferenças significativas relativamente ao consumo de alguns alimentos como o gelado, os cereais e as bolachas tipo Maria.A exposição ao glifosato é preocupante, principalmente tendo em conta as idades dosparticipantes e a toxicidade do herbicida. Afigura-se necessário realizar estudos adicionais com maior número de amostras e maior diversidade geográfica.
The use of glyphosate has been increasing over the years, making it one of the most consumed herbicides in the world. Its effects on the human organism and on the environment have been analyzed and are of concern, as exposure to the herbicide increases.The aim of the present study was to determine glyphosate in the urine of children from different regions of Portugal, including urban and rural areas, to identify levels of exposure and to relate to dietary patterns.Of the 41 children, 46.34 % (n = 19) were male, aged between 2 and 12 years old, and 53.66 % (n = 22) were female, aged between 2 and 13 years old. The collection of samples (first urine of the day) occurred between July 2018 and February 2019. In addition to the consent form, a questionnaire was filled by the children's parents, with anthropometric characteristics, eating habits, lifestyle and use of pesticides. Glyphosate was analyzed using the ELISA method (LOD = 0.6 µg/L) and was detected in 95.12 % of the samples, with mean values of 1.85 ± 0.82 µg/L. and minimum and maximum values of 0.87 and 4.35 µg/L, respectively.Of the 13 most recent studies that determined glyphosate in urine, only 4 used samples from children, 3 of which showed higher values than the present study. However, only 1 of the cases had a higher incidence than that mentioned in this study.The differences between the glyphosate concentrations in the urine of children aged 3 years or less (1.20 ± 0.32 µg/L), compared to the concentrations found in children between 7 and 9 years (2.12 ± 1.00 µg/L) were close to statistical significance (p = 0.0631), and can be justified by the increased activity of the enzyme PON1, at 7 years. Higher levels of contamination were found in children living near agricultural areas (<1 km, 2.0 ± 0.97 µg/L), with a higher percentage of consumption of homemade foods (75-100%, 2.72 ± 0.45 µg/L) and whose parents applied herbicides in the yard (2.5 ± 1.17 µg/L), when compared to the garden (1.4 ± 0.43 µg/L). In addition, there were significant differences regarding the consumption of some foods such as ice cream, cereals and Maria-like cookies.Exposure to glyphosate is worrying, especially considering the ages of the participants and the toxicity of the herbicide. It appears necessary to carry out additional studies with a larger number of samples and greater geographical diversity.
