dc.description.abstractA Doença de Alzheimer é a forma mais comum de demência no hemisfério ocidental, que se caracteriza por sintomas como a perda de memória, dificuldades no discurso, perda da capacidade de reconhecer objectos e pessoas, e incapacidade de realizar actividades da vida diária. Trata-se de uma doença neurodegenerativa, que tem vindo a ser intensivamente estudada nas últimas três décadas. Apesar de, inicialmente, se pensar que teria um carácter genético, os números indicam que só 5 a 10% dos casos são de origem inteiramente genética e, habitualmente, manifestam-se antes dos 65 anos. Como tal, os restantes 90 a 95% são casos em que a genética desempenha um papel pouco relevante. Nos últimos anos houve uma mudança do paradigma em relação à verdadeira origem para o desenvolvimento desta doença. Vários estudos permitiram identificar múltiplos mecanismos, desde genéticos, peptídeos β-amilóide, stresse oxidativo e inflamação que, de modo interactivo poderão estar na génese da doença de Alzheimer. Mais recentemente, a revisão dos resultados de publicações mais antigas, juntamente com as investigações realizadas recentemente, apontam para que a Doença de Alzheimer possa ser, na verdade, uma condição metabólica inerente a um aumento da resistência à insulina, i.e., um tipo de diabetes – diabetes tipo 3. Deste modo, são cada vez mais os investigadores que se debruçaram a compreender melhor esta patologia deste novo ponto de vista, assim como a encontrar novas estratégias de tratamento. dc.description.abstractAlzheimer’s Disease is the most common form of dementia in the western hemisphere. It is a neurodegenerative condition that has been intensively studied for the past three decades. Although initially was though that it could be a genetic disorder, the numbers indicate that only 5 to 10% of the cases have genetic origins and usually they manifest prior to the age of 65, whereas in 90 to 95% of the cases the genetics factor is close to irrelevant. Over the last years there has been a paradigm shift related to the real origin for the development of the disease. Intensive research has identified multiple interactive and parallel mechanisms, ranging from genomics, amyloid-β peptides, oxidative and infllmatory factors, likely causative of the development of Alzheimer´s disease. However, the review of older publications and the current investigations point that Alzheimer’s Disease may be, in fact, a metabolic condition inherent to an increase in insulin resistance i.e., a form of diabetes – Diabetes type 3. Hence, there is a growing number of researchers that struggle to better understand this disease, as well as to find new treatment strategies.
