dc.description.abstract | O Síndrome da Imunodeficiência Adquirida (SIDA), decorrente da infeção pelo Vírus da Imunodeficiência Humana (VIH), é uma condição clínica potencialmente fatal que persiste mundialmente como um drástico problema de saúde. Desde que foi associado ao processo de adsorção pelo VIH, vários estudos têm sido desenvolvidos com o objetivo de identificar novas moléculas terapêuticas capazes de atuar no Recetor de Quimiocinas CCR5, tendo sido o Maraviroc®, em 2007, o primeiro medicamento aprovado como efetivo inibidor terapêutico. Posteriormente, a aparente cura de um doente infetado com HIV - "O Paciente de Berlim" -, após ter sido transplantado com células hematopoiéticas desprovidas da expressão do CCR5, proporcionou o interesse no co-desenvolvimento de múltiplas técnicas de terapia génica. Considerando as limitações existentes no atual protocolo de tratamento anti-retroviral e a inexistência de uma vacina aprovada, a capacidade das Nucleases de Design modificarem genomas complexos desencadeou um notável impacto na área da biotecnologia. Nos últimos anos, a tecnologia de edição génica pela ZFN, TALEN and CRISPR/Cas9 surgiu como uma solução promissora capaz de mimetizar a ocorrência natural da mutação CCR5/Δ32 e, em consequência, garantir consistentemente que o CCR5 não se expressa à superfície das células-alvo do VIH, permitindo uma contínua resistência à entrada do vírus. dc.description.abstract | Acquired Immunodeficiency Syndrome (AIDS), caused by Human Immunodeficiency virus (HIV), is a life-threatening disorder that persists worldwide as a severe health problem. Since it was connected with HIV attachment process, intensive studies lead to the development of small molecular drugs targeting CCR5 with Maraviroc® becoming, in 2007, the first clinically approved chemokine receptor inhibitor. Latterly, the apparent HIV cure in a patient transplanted with CCR5-depleted hematopoietic stem cells - “The Berlin Patient” - has led to a resurgence of interest in inactivating CCR5 through gene therapy, either as a stand-alone approach or as an adjuvant to pharmacological drug regimens. Given the shortcomings of the current anti-retroviral treatment procedure and the non-availability of a licensed vaccine, the aptitude to modify complex genomes with Designer Nucleases has had a noteworthy impact on biotechnology. Over the last years, ZFN, TALEN and CRISPR/Cas9 gene-editing technology has appeared as a promising solution that mimics the natural occurring CCR5/Δ32 mutation and then permanently guarantees the absence of CCR5-expression on HIV target-cells surface, leading to a continuous resistance to the virus entry and, ultimately, proving that cellular immunization from infection could be, in fact, a conceivable therapeutic approach to finally achieve the long-awaited HIV functional cure. | eng |
