dc.description.abstractO interesse na exploração das algas nas mais variadas áreas (como farmacêutica, nutracêutica, alimentação e cosmética) tem vindo a aumentar uma vez que são produtos naturais com variados potenciais naturais ainda pouco estudados. Deste modo, o seu estudo e dos seus compostos torna-se relevante.Com este trabalho, foi avaliado o melhor método para o fracionamento e extração de compostos da alga vermelha Grateloupia turuturu, tendo em conta as gerações do seu ciclo de vida. Foi ainda avaliado o potencial antibacteriano e antifúngico dos vários extratos obtidos por diferentes solventes. E, por fim, foram analisadas em Fourier transform infraref spectroscopy- attenuated total reflectance (FTIR-ATR) amostras de alga seca, de modo a identificar alguns compostos presentes, assim como melhor caracterização dos seus polissacarídeos.O método que nos propusemos otimizar foi o método de extração sequencial previamente descrito e utilizado, mas não especificamente para a alga em estudo. Para a extração dos compostos recorreu-se à utilização de vários solventes, nomeadamente n-hexano, etanol absoluto e água. Para o estudo da atividade antibacteriana e antifúngica, apenas foram testados os extratos de etanol e polissacarídeos. É de notar que foram feitos extratos para duas gerações do ciclo de vida da alga, geração tetrasporófita e geração gametófita frutificada. A capacidade antibacteriana dos extratos foi testada contra Escherichia coli e Staphylococcus aureus e a capacidade antifúngica foi testada contra Candida albicans. Os testes realizados tiveram por base o método de microdiluição e o ensaio de citotoxicidade foi feito com recurso à utilização de Iodonitrotetrazolium chloride (INT). Na análise em FTIR-ATR foi utilizado o equipamento IFS 55 spectrometer, using a Golden Gate single reflection Diamond ATR system e a leitura foi feita nos comprimentos de 500 a 1500 com posterior identificação dos picos mais relevantes, também foram comparadas as gerações do ciclo de vida da alga.Após otimização do método de extração sequencial, foi através do protocolo C que obtivemos extratos com maior quantidade e estéreis. Nenhum extrato revelou capacidade antibacteriana e antifúngica total à concentração máxima usada (12,5mg/mL). Porém o extrato de etanol mostrou ser capaz de inibir o crescimento de E. coli e S. aureus significativamente (32,7% e 22,5%, respetivamente), os extratos de polissacarídeos por sua vez mostraram efeito inibitório significativo sobre E. coli à mesma concentração (67,7%), contudo sem qualquer inibição sobre S. aureus. Para C. albicans verificou-se também efeito inibitório ligeiro (12.7%) não tendo, porém, significado estatístico.Através das análises em FTIR-ATR observou-se que G. turuturu é composta maioritariamente por ésteres de sulfato e carragenana kappa, relativamente às diferenças entre gerações estas não são significativas sendo maioritariamente causada pela quantidade de agar.Com este trabalho fizemos otimização do método de extração sequencial de compostos biativos de G. turuturu, verificámos que os extratos de etanol podem ser promissores na inibição do crescimento de estirpes bacterianas, como E. coli e S. aureus, que os extratos de polissacarídeos podem também ter relevância contra estirpes bacterianas como E. coli e o potencial antifúngico pode ser ainda mais explorado. Relativamente à composição os compostos polares parecem deter as propriedades antibacterianas mais relevantes, a nível dos polissacarídeos as carragenanas podem deter potencial antinfúngico, sendo também importante e relevante explorar a carragenana kappa, esteres de sulfato e a sua composição rica em agar, dados corroborados pela análise de compostos por FTIR-ATR.por
dc.description.abstractThe interest in the exploitation of algae in the most varied areas (such as pharmaceuticals, nutraceuticals, food and cosmetics) has been increasing since they are natural products with varied natural potentials that have not yet been studied. In this way, your study and its compounds become relevant.With this work, we evaluated the best method for the fractionation and extraction of compounds from the red algae Grateloupia turuturu, considering the generations of its life cycle. It was also evaluated the antibacterial and antifungal potential of the various extracts obtained by different solvents. Finally, we analyzed Fourier transform infraref spectroscopy-attenuated total reflectance (FTIR-ATR) samples of dry algae to identify some compounds present, as well as better characterization of their polysaccharides.The method we proposed to optimize was the sequential extraction method previously described and used, but not specifically for the alga under study. For the extraction of the compounds the use of various solvents, namely n-hexane, absolute ethanol and water was used. For the study of antibacterial and antifungal activity, only the extracts of ethanol and polysaccharides were tested. It should be noted that extracts were made for two generations of the algae life cycle, tetrasporophyte generation and fructified gametophyte generation. The antibacterial capacity of the extracts was tested against Escherichia coli and Staphylococcus aureus and the antifungal capacity was tested against Candida albicans. The tests performed were based on the microdilution method and the cytotoxicity assay was performed using Iodonitrotetrazolium chloride (INT). For C. albicans, there was also a slight inhibitory effect (12.7%), but it did not have a statistical significance.It was observed that G. turuturu is mainly composed of sulfate esters and kappa carrageenan, in relation to the differences between generations, these are not significant, being mainly caused by the amount of agar.With this work we have done an optimization of the sequential extraction method of biative compounds of G. turuturu, we have verified that the ethanol extracts can be promising in inhibiting the growth of bacterial strains, such as E. coli and S. aureus, that the extracts of polysaccharides can also be relevant against bacterial strains such as E. coli and the antifungal potential may be further explored. Regarding the composition the polar compounds seem to have the most relevant antibacterial properties, carrageenans may have anti-fungal potential in the polysaccharides, and it is also important and relevant to exploit kappa carrageenan, sulfate esters and their agar-rich composition, as corroborated by the analysis of compounds by FTIR-ATR.eng
