Abstract: Os estrogénios são potentes neuroprotetores tanto in vitro como in vivo em modelos de neurotoxicidade. Num modelo in vivo de neuroprotecção, isquemia cerebral transitória, os estrogénios foram ativos na proteção do tecido cerebral contra esta lesão pro-oxidante. Apesar de grande parte dos efeitos dos estrogénios no cérebro serem mediados através de recetores de estrogénio (ERs), evidências mostram que a neuroprotecção também pode ser mediada por mecanismo que não envolvem o ER. Os estrogénios provavelmente atuam através do ciclo redutivo que envolve a glutationa e a HMS. Recentes estudos sugerem que os estrogénios são capazes de potenciar o potencial redutivo do HMS, e a redução do GSSH em GSH através do NADPH-induzido. Pequenas modificações estruturais no 17β- estradiol podem eliminar parcialmente ou mesmo totalmente a ligação aos ERs. Por exemplo o 17α-estradiol (17α-E2), diastereómero do 17β-E2, que é produzido naturalmente, apresenta consideravelmente menos afinidade para os recetores ERα e Erβ. Estudos de estrutura atividade revelaram que modificações no anel A são as mais prováveis de aumentar o efeito neuroprotetor, no entanto modificações no anel B e C também podem ser eficientes. Partindo do trabalho já existente de novas reações e processos para obtenção de esteróides bioativos, foram exploradas modificações seletivas de posições-chave do esqueleto de estradiol para preparar novos derivados.Diversas estratégias sintéticas foram usadas para modificar a estrutura do estradiol tanto no anel A como B.
Estrogens are potent and efficient neuroprotectives both in vitro and in vivo in several models of neurotoxicity. In an in vivo model for neuroprotection, transient cerebral ischemia, estrogens were active in protection of brain tissue from this pro-oxidant insult. Although many of the effects of estrogens in the brain are mediated through estrogen receptors (ERs), there is evidence that neuroprotective activity of estrogens can be mediated by non-ER mechanisms. The estrogens are likely to work through a redox cycle that involves glutathione and the HMS. Recent studies suggest they are able to tap the large reducing potential of the HMS, and through NADPH-induced reduction of GSSH to GSH, and able to apply this reducing potential to lipid membranes using estrogens as a mediator. Minor modifications to the structure of 17β-E2 can partially or completely eliminate ER-binding. For example, 17α-estradiol (17α-E2), a diastereomer of 17β-E2 produced naturally in ungulates binds to both ERα and ERβ with a 40-fold lower affinity than 17β-E2 .Previous studies showed that, modifications on the A ring are the most likely to enhance the neuroprotection, although modifications on rings B and C can also be efficient. Within our framework of new reactions and processes towards bioactive steroids we have been exploring the selective modification of key positions of the estradiol scaffold to prepare new derivatives.Diverse synthetic strategies were explored to modify the structure of estradiol rings A and B.
