dc.description.abstract | O aumento das resistências aos antibióticos tem motivado a necessidade de procurar estratégias antibacterianas alternativas. Os bacteriófagos, vírus que infetam bactérias, foram descobertos no início do século XX e foram utilizados com sucesso no tratamento de infeções bacterianas durante algumas décadas. Contudo, o interesse na terapia fágica diminuiu depois da descoberta dos antibióticos. Atualmente, os fagos são novamente apontados como uma possível opção e várias vantagens sobre os antibióticos são já reconhecidas. Apesar disso, existem ainda alguns desafios a superar de forma a que a terapia fágica possa ser utilizada rotineiramente. Uma solução para ultrapassar algumas das limitações desta terapêutica poderá ser a melhoria da terapia fágica convencional –administração de fagos modificados, enzimas fágicas ou combinação com antibióticos. Os ensaios clínicos realizados indicam que a terapia fágica é segura e eficaz. Além disso, já existem produtos aprovados para aplicações relacionadas com a segurança alimentar e vários ensaios clínicos de fase II e III estão atualmente a decorrer. dc.description.abstract | The rise in antibiotic resistance has prompted the need for alternative antibacterial strategies. Bacteriophages, viruses that infect bacteria, were discovered in the beginning of the 20th century and were successfully employed to treat bacterial infections during a few decades. However, interest in phage therapy decreased after the discovery of antibiotics. Nowadays, phages are once again regarded as a possible option and several advantages over antibiotics are already recognized. Despite this, there are still some challenges to overcome in order for phage therapy to be routinely used. One solution to surpass some of the limitations of this therapy could be the improvement of conventional phage therapy –administration of modified phages, phage enzymes or combination with antibiotics. Conducted clinical trials suggest that phage therapy is safe and effective. Furthermore, there are already phage related products approved for food safety applications and several phase II and III clinical trials are currently ongoing.
