Abstract: Desde sempre que os produtos naturais têm vindo a exercer um papel importante tanto na medicina tradicional como na indústria farmacêutica, constituindo a base para novos compostos biologicamente ativos com interesse terapêutico. Os terpenos integram umas das mais vastas e interessantes classes de produtos naturais encontrados principalmente nas plantas. Entre eles, destacam-se os diterpenos do tipo abietano que são compostos que têm despertado grande interesse na comunidade farmacêutica.Nos últimos anos, estudos intensivos realizados nos diterpenos do tipo abietano revelaram novos compostos desta família de produtos naturais. O Ácido carnósico é um dos diterpenos do género abietano aromático mais relevantes, é principalmente encontrado e extraído do Rosmarinus Officinalis L.. Este diterpeno é reconhecido pela sua potente atividade antioxidante e tem sido muito explorado na indústria alimentar e cosmética. Além disso, o Ácido carnósico tem sido amplamente estudado nos últimos anos devido as suas atividades biológicas, nomeadamente a atividade anticancerígena, antifúngica e neuro protetora. Considerando o potencial biológico do Ácido carnósico, estudos anteriores que descreveram a síntese de derivados semissintéticos relataram uma melhoria na sua atividade biológica. Esses estudos sugerem que modificações químicas na estrutura do Ácido carnósico podem contribuir para melhorar suas propriedades farmacológicas. O objetivo deste trabalho, passou pelo design, síntese e purificação de alguns derivados semissintéticos do Ácido carnósico, com posterior avaliação da sua pureza e elucidação estrutural através do recurso a técnicas analíticas, tais como, espectroscopia de ressonância magnética nuclear e ponto de fusão.
For a long time, natural products have been playing an important role not only in traditional medicine but also in the pharmaceutical industry, as the basis of new bioactive compounds of interest. Terpenes represent one of the most diverse and interesting classes of natural products mainly found in plants. Among them, abietane type diterpenes, have attracted growing attention from the pharmaceutical community. In the past few years, intensive studies on abietane type diterpenes have revealed many new members of this family of natural products. carnosic acid is one of the most relevant aromatic abietane type diterpenes and is mainly found and extracted from Rosmarinus officinalis L.. This diterpene is recognized for its potent antioxidant activity, being widely explored within the food and cosmetic industry. Furthermore, carnosic acid has been extensively studied over the last few years for its biological properties, namely anticancer, antifungal and neuroprotective activities. Considering its biological potential, previous studies reported the synthesis of semisynthetic derivatives of carnosic acid with improved biological activity. These studies suggested that chemical modifications of the structure of carnosic acid may contribute to improve its pharmacological properties.The aim of this work was the design, synthesis and purification of some semisynthetic derivatives of carnosic acid, with posterior evaluation of their purity and structural elucidation through the use of some analytical techniques such as nuclear magnetic resonance spectroscopy and melting point analysis.
