Abstract: A pele cobre toda a extensão do corpo e é um órgão de relativa complexidade, que tem como função principal atuar como uma barreira protetora entre o meio exterior e os órgãos e tecidos internos. É facto que está diariamente exposta e portanto é um alvo relativamente fácil de danificar e a sua integridade pode ser afetada por fatores como disfunções genéticas, traumas agudos, feridas crónicas ou intervenções cirúrgicas, sendo a razão mais comum os traumas térmicos. O tratamento passa portanto por revestir as feridas da melhor forma e mais adequada possível para promover a sua cicatrização e alcançar a cura.Existem já diversos revestimentos de feridas, no entanto há ainda muitos materiais e métodos a explorar, pelo que os cientistas continuam a investigar novos sistemas ou a melhorar os já existentes. Face ao referido, uma solução passa pela Engenharia de Tecidos e pela formulação de membranas que possam ser aplicadas como revestimentos ideais de feridas.O foco do presente trabalho foi o desenvolvimento de membranas assimétricas eletrofiadas para a regeneração de tecidos, usando materiais que possam fornecer as condições necessárias para fixação e proliferação celular, mimetizando as funções naturais da pele conduzindo à sua regeneração. Foram desenvolvidas membranas assimétricas fibrosas, onde a camada base é composta por policaprolactona (PCL) e poliácido láctico (PLA) e a segunda camada por diferentes formulações de PCL e gelatina funcionalizada com Anidrido Metacrílico (MAA), para torna-la fotoreticulável. Estas foram então produzidas por diferentes abordagens e metodologias da técnica Electrospinning. Na primeira abordagem foram produzidas as camadas base por electrospinning de mistura. Na segunda e terceira abordagem foram respetivamente produzidas as membranas assimétricas, por electrospinning de mistura e coaxial. Através do electrospinning coaxial pretendeu-se produzir fibras com uma estrutura do tipo núcleo e casca, em que o núcleo é constituído por PCL e a casca por GelMA.As membranas assimétricas desenvolvidas foram então fotoreticuladas por irradiação de luz UV, reticulando a gelatina previamente funcionalizada, posteriormente foram caracterizadas quanto às suas propriedades químicas, morfológicas e biológicas. O recurso ao ATR-FTIR permitiu identificar a presença das bandas típicas da gelatina e da PCL na segunda camada da membrana assimétrica. O teste da perda de massa revelou o sucesso da fotoreticulação, visto que esta foi menos evidente nas membranas fotoreticuladas que nas suas percursoras originais. Os ângulos de contacto dinâmicos foram também avaliados e mostraram que a membrana base é marcadamente hidrofóbica, por oposição às restantes que exibem no geral um carácter hidrofílico.A estrutura da superfície das membranas é, como se esperava, desorganizada devido às técnicas pelas quais foram produzidas, como se comprova pela análise das imagens obtidas por SEM. Esta análise permitiu também avaliar a influência do teor de GelMA nas fibras, pois quanto mais elevada é a quantidade de PCL mais fundido é o aspeto das fibras. Além disso, revelou também que por electrospinning de mistura os diâmetros das fibras é menor que os das suas homólogas produzidas por electrospinning coaxial.Os testes de hemocompatibilidade revelaram que todas as membranas induzem a formação de trombos, sendo que esta foi mais notória na membrana com 100 % GelMA. A dissolução da gelatina nesta membrana fez com que a camada base constituída por polímeros hidrofóbicos promovesse a formação dos trombos. Segundo os resultados obtidos, as membranas mais trombogénica são as produzidas por electrospinning coaxial. Quanto ao índice hemolítico, apenas a membrana com 100 % GelMA apresenta um carácter ligeiramente hemolítico, sendo as restantes não hemolíticas.Finalmente os estudos de biocompatibilidade demonstraram que as membranas assimétricas produzidas promoveram uma boa adesão e proliferação dos fibroblastos dermais. No que toca à atividade antimicrobiana os resultados não foram de todo satisfatórias, visto que as membranas não inibiram a atividade da bactéria à qual tiveram contacto, deixando-a proliferar. Este problema pode ser futuramente contornado com a adição de um antibiótico.Em suma, ainda que seja necessária uma análise mais aprofundada, os resultados fazem crer que as membranas desenvolvidas neste trabalho são boas candidatas para a aplicação pretendida.
Skin covers the whole extension of the human body and it’s an organ of relative complexity, whose biggest function is to act as a protective barrier between the outer environment and the internal organs and tissues. It is a fact that skin is exposed every day and therefore is easily damaged and that its integrity could be affected by several stress factors like genetic anomalies, acute trauma, chronic wounds or surgical interventions, with the more common one being thermal trauma. The treatment therefore involves coating the wound in the most suitable way to trigger the healing process so that a full recovery can be achieved. There is already a lot of diversity in wound dressings, however there are still several materials and methods yet to be explored, and scientists are still researching new systems or improving the already existing ones. Due to the described above, one solution may involve Tissue Engineering and the use of membranes which can be applied as ideal wound dressings. The focus of this work was the development of asymmetrical electrospun membranes for tissue regeneration, using materials that could provide the required conditions for cell attachment and proliferation, mimicking the skin’s natural functions leading to its regeneration. Asymmetrical fibrous membranes were developed, in which the base layer is composed of polycaprolactone (PCL) and polylactic acid (PLA), and the second layer is composed of PCL and gelatin functionalized with methacrylic anhydride (MAA), so that it could be photopolymerized. These membranes were produced via distinct approaches and methodologies of the electrospinning technique. In the first approach the base layers were synthesized by blending electrospinning and at the same time the gelatin was modified. In the second and third approaches, the asymmetrical membranes were produced respectively by blending and coaxial electrospinning. The main goal for the use of coaxial Electrospinning was to produce fibers with core and shell structure, in which the core is composed of PCL and the shell of GelMA. The asymmetrical membranes were then photocrosslinked by UV light irradiation, crosslinking the previously modified gelatin. Afterwards, all the membranes were characterized by their chemical, morphological and biological properties.The ATR-FTIR analysis made it possible to identify the presence of both polymers used on the second layer of the asymmetrical membrane composition. The weight loss study demonstrated the success of the photocrosslinking process, evidenced by the fact that it was less obvious in the photocrosslinked membranes when compared to their original precursors. The dynamic water contact angle measurements were also assessed and they evidenced the hydrophobic character of the layer base, as opposed to the others that exhibit a hydrophilic character. As expected, the SEM analysis showed that the surface structure of the membranes is disorganized, due to the technique used to produce them. Through this analysis the influence of the GelMA ratio on the fibers was appraised as well, showing that the the higher the percentage of PCL the more melted the fibers will appear. Besides that, it was revealed that the fiber diameters are bigger in the membranes produced by blending electrospinning than their homologous ones produced by coaxial electrospinning.The hemocompatibility tests revealed that all membranes are thrombogenic, with higher intensity in the membrane with 100% GelMA. The dissolution of the gelatin in this membrane increased thrombogenecity allowing exposure of the base layer of hydrophobic polymer. According to the results, coaxial electrospinning produces membranes with higher thrombogenecity. As for the hemolytic index, only the membrane with 100% GelMA revealed a slightly hemolytic character, the remaining ones being non-hemolytic.Finally, the biocompatibility studies revealed that the asymmetrical membranes promoted a good attachment and proliferation of dermal fibroblasts. With respect to antimicrobial activity, the results showed that the materials were not bactericidal, since the membranes did not inhibit the activity of the bacteria they came in contact with, instead allowing them to proliferate. This problem could be worked around in the future with the addition of an antibiotic.In summary, even through a more in-depth analysis is required, the results suggest that the membranes developed are potential candidates for the intended application.
