Title: Envelhecimento e nutrição : contracção calórica e longevidade

Abstract: O processo de envelhecimento, que envolve a acumulação de diversas alterações ao nível genético e celular, e um desequilíbrio da balança homeostática, que conduz ao aparecimento de patologias crónicas e em último caso à morte, sempre foi alvo de interesse por partedo Homem. Existem diversas teorias sugeridas para o explicar. O objectivo deste trabalho passa por tentar entender os mecanismos deste processo e os factores que o influenciam, para a definição de uma possível estratégia de envelhecimento saudável. A metodologia passa por uma revisão da literatura mais recente sobre este assunto. A contracção calórica, a única estratégia não genética capaz deaumentar a longevidade em diversas espécies, é alvo de enorme investigação. Os mecanismos celulares e moleculares responsáveis pelos efeitos induzidos pela contracção calórica ainda não estão totalmente esclarecidos e requerem maior investigação. Estudos em humanos são de difícil realização e só agora começam a mostrar resultados mais conclusivos, embora, até ao momento, só tenha sido possível executar ensaios de curta duração, estando neste momento em andamento um estudo de longo termo. Os resultados são promissores na modificação de alguns biomarcadores relacionados com a idade, mas mais estudos são necessários para avaliar o real efeito sobre a longevidade e sobre a saúde. Os miméticos apresentam-se com boas perspectivas futuras para obviar alguns dos problemas da implementação de programas de longa duração de contracção calórica. A implementação de protocolos de longa duraçãode contracção calórica não parece actualmente uma medida exequível pelas dificuldades inerentes, quer de motivação, culturais ou outras. É necessário comparar os efeitos benéficos desta intervenção nutricional, com aqueles relacionados com hábitos alimentares saudáveis, bem como maior investigação sobre esta intervenção dietética a longo prazo em humanos.
The aging process, which involves the accumulation of several changes in genetic and cellular levels, and an unbalanced homeostasis, leading to the emergence of chronic diseases and ultimately death, always been the target of interest on the part of Humankind. There are various theories suggested to explain it. The aim of this work involves trying to understand the mechanisms of this process and the factors that influence it, to define a possible strategy for healthy aging. The methodology involves a review of recent literature on this subject.The caloric restriction, the only non-genetic strategy that seems to increase longevity in several species, is subject of huge research. The cellular and molecular mechanisms responsible for the effects induced by caloric restriction are not yet fully understood and require further investigation. Human studies are difficult to perform and only now beginning to show more conclusive results, although it has only been possible to perform short-term trials, and currently is in progress a long-term study. The results are promising for the modification of some age-related biomarkers, but more studies are needed to assess the real effect on the longevity and health. The mimetics are presented with good prospects to overcome some of the problems of implementing programs of long-term caloric contraction.The implementation of protocols of long-term caloric restricton does not seem to be achievable, by the difficulties inherent in either of motivation, cultural or other.Moreover, it is necessary to compare the beneficial effects of this nutritional intervention, with those related to healthy eating habits, and further research about this long-term dietary intervention in humans.
