Abstract: Apresentando-se como uma das grandes epidemias do século XXI, a obesidade constitui um grave, actual e exponencial problema de Saúde Pública. O século XX constituiu um marco no interesse médico relativamente à obesidade, com um número crescente de descobertas relacionadas com os seus malefícios que contribuíram para se deixar de considerar a mesma não uma condição mas sim uma doença com graves consequências médicas, psicológicas bem como sociais quando não tratada. Surgem assim diversos estudos centrados nos mecanismos de acção de possíveis fármacos com vista a um objectivo comum, a redução do peso corporal. Ao longo dos anos, foram vários os fármacos que elevaram as expectativas da comunidade científica sem, contudo, apresentarem relação eficácia/segurança satisfatória. O orlistat é actualmente o único fármaco aprovado no que diz respeito à terapêutica médica farmacológica da obesidade e, apesar da multiplicidade de estudos em curso acerca de novos alvos terapêuticos nesta área, são escassos os que apresentam resultados favoráveis.Pretende-se realizar um artigo de revisão que permita conceder uma perspectiva histórica dos fármacos que fizeram parte da terapêutica da obesidade e as razões pelas quais foram abandonados. Pretende-se, igualmente, analisar os que existem actualmente no mercado e, ainda, perceber quais são as perspectivas futuras de que dispomos.
Presenting itself as one of the major epidemics of the 21st century, obesity is a serious, current and exponential problem of public health. The 20th century was a milestone in medical interest regarding obesity, with an increasing number of discoveries related to its harmful effects that contributed to no longer consider it a condition but a disease with serious consequences, medical, psychological and social consequences whenever left untreated. So, several studies arise focusing on possible mechanisms of action of drugs with a view to a common goal, the reduction of body weight. Over the years, several drugs raised up the expectations of the scientific community without, however, have shown efficacy/safety satisfactory relationship. Orlistat is currently the only drug approved regarding medical therapeutic pharmacological obesity and despite the multitude of ongoing studies about new therapeutic targets in this area, there is little that presents favorable results. The intention is then to conduct a review article that will grant a historical perspective of the drugs that were part of the treatment of obesity and the reasons why they were abandoned. It is also intended to analyze what drugs currently exist on the market and still realize what future prospects are now available.
