dc.description.abstractO piruvato é um substracto energético com reconhecidas propriedades cardioprotectoras. Sabe-se hoje que isso se deve não só à sua actividade antioxidante, mas também ao facto de reduzir a acidose intracelular, modular a cinética intracitosólica do cálcio e melhorar a contractilidade cardiomiocitária. Porém, só recentemente é que se começou a compreender a importância das mitocôndrias cardíacas para a ocorrência destas alterações benéficas, sendo que os mecanismos concretos de actuação do piruvato a nível mitocondrial estão ainda largamente por esclarecer. Com o objectivo de estudar o efeito do piruvato na função mitocondrial de corações isquémicos utilizou-se um modelo animal de perfusão ex-vivo com um sistema de Langendorff, seguido de isquemia na presença ou ausência de piruvato. Avaliou-se o potencial eléctrico da membrana mitocondrial, o consumo de oxigénio (O2) pela cadeia respiratória (e o respectivo índice de controlo respiratório) e as cargas energéticas geradas com diversos substratos energéticos. Verificou-se que o piruvato actua no sistema oxidativo, melhorando de forma não significativa o consumo de O2 durante a isquemia (elevação do índice de controlo respiratório – ICR), mas sobretudo ao nível do sistema fosforilativo, encurtando de forma significativa o ciclo fosforilativo (Lag phase) e melhorando a produção de ATP (aumento da carga energética), o que permitiu assegurar uma maior integridade das estruturas membranares mitocondriais, com manutenção do potencial eléctrico após o ciclo fosforilativo. Estes achados permitem compreender melhor os mecanismos inerentes aos efeitos citoprotectores do piruvato em portadores de cardiopatia isquémica, demonstrando claramente a importância das mitocôndrias cardíacas neste processo.en_US
dc.description.abstractPyruvate is an energy substrate with known cardioprotective activity. We know now that this is due not only to its antioxidant activity, but also to its reduction of intracellular acidosis, modulation of intracytosolic calcium and improvement of cardiomyocyte contractility. However, the role of cardiac mitochondria in such positive effects has only recently begun to be understood and the exact mechanisms of the effect of pyruvate on mitochondria are still largely unknown. Aiming to study the effect of pyruvate on cardiac mitochondrial function during acute ischemia, we used an ex-vivo animal model, perfused in a Langendorff system and then subjected to ischemia in the presence and absence of pyruvate. We evaluated the mitochondrial membrane electrical potential, the respiratory chain O2 consumption (and respiratory control ratio) and the energy charges generated with different energy substrates. We conclude that pyruvate has some effect on the mitochondrial oxidative system (by non-significantly improving the respiratory control ratio), but its main action is on the phosphorylation system, significantly decreasing the time taken to complete a phosphorylation cycle (lag phase) and improving ATP production (increase in energy charge), thus allowing better maintenance of mitochondrial membrane structure, with consequent improvement of the electrical potential after a phosphorylation cycle. These findings have enabled better understanding of the mechanisms behind pyruvate cytoprotection in ischemic cardiomyopathy, clearly highlighting the essential role of cardiac mitochondria in this process.-
